    Porcine parvovirus 4 (PPV4) has been reported in several countries and the high rate of concurrent PCV2 infection with PPV4 may trigger the “porcine circovirus disease” (PCVD). This has awakened the interest in how PPV4 virus behaves. The aim of this study was to show that, like other swine viruses, the elimination of PPV4 in semen is intermittent. The study was conducted in the Unidade de Pesquisa e Desenvolvimento de Itapeva da Agência Paulista de Tecnologia dos Agronegócios Regional (Apta Regional). Over a period of six months, four boars were monitored and the semen was collected every 10 days, totalizing 74 semen samples. Through PCR testing, PPV4 was detected in 32/74 (43.2%) semen samples. Three sequences of 284 nt, showed identity varying from 98.2% to 100%, confirming the detection of the virus in all boars. Semen volume and sperm motility did not indicate significant difference between PPV4 positive or negative semen samples (p > 0.05). There was no significant difference between the monthly distributions of positive and negative samples. This is the first study showing intermittent shedding of PPV4 in naturally infected boars. Absence of typical clinical signs and/or influence in semen quantity was also observed.

    Bovine vaccinia (BV) is a vesicular disease induced by the Vaccinia virus (VACV) that affects milk production and is an occupational zoonosis. This research had the following objectives: (i) detection of VACV by qPCR in cattle with clinical suspicion of vesicular disease; (ii) symptoms characterization in animals and milkers with clinical suspicion of the disease and virus detection in humans; and (iii) identification of risk factors for infections of VACV in herds from several Brazilian states. A total of 471 bovine epithelial samples from dairy farms, in 15 Brazilian states, were evaluated between 2007 and 2012. The samples were tested by quantitative PCR (qPCR) using SYBR Green® reagents, validated with a lower limit of detection of 100TCID50/50µL (1.7x100 viral particles), and 45.1% of VACV positive samples were detected. Using official forms for epidemiological investigation (FORM-IN), the risk factors for VACV infections in cattle were determined to be farms with a lack of technological facilities (P= 0.029) and the presence of rodents (P= 0.001). There was an effect of seasonality in cattle with a higher occurrence of BV during the dry season. A total of 420 epidemiological questionnaires were applied at public health care centers, where 100% of the milkers had vesicular lesions on their hands (98.1%) and on their arms (6.9%). The most frequent clinical symptoms in humans were: local swelling (74.2%), headache (20.7%), fever (10.4%) and inguinal lymphadenopathy (74.2%). Only 19.98% of milkers aged between 39 and 58 years were seroreactive to VACV and were immunized with the human anti-smallpox vaccine. There was an increase in the frequency of BV in older individuals due to their natural decrease in specific immunity. It has been shown that the implementation of zootechnical management techniques and health planning are important for the prevention of BV in animals and humans.

    O plasma sanguíneo em pó (PSP), produto natural de indústria frigorífica, tem mostrado efeitos benéficos sobre o crescimento e desempenho de leitões desmamados precocemente. Atualmente, embora o circovírus suíno 2 (PCV2) tenha grande importância para a suinocultura, não há informações sobre o impacto do uso de PSP e a resposta imune ao PCV2 em infecções naturais. Este trabalho avaliou diferentes níveis de inclusão de PSP em dietas de leitões e as cargas virais de PCV2 correspondentes. Quatro níveis de inclusão de PSP foram testados em dois períodos consecutivos: 0, 2, 4 ou 6% durante o período 1 (14 aos 28 dias de idade) e 1, 2 ou 3% de PSP durante o período 2 (29 a 42 dias de idade). No período 3 (42 aos 56 dias de idade), todos os leitões foram alimentados com dieta isenta de PSP. Amostras de soro foram coletadas semanalmente e testadas para anticorpos anti-PCV2 e carga de DNA de PCV2. As concentrações de 6% e 3% de PSP fornecidas nas rações durante o período 1 e 2, respectivamente, influenciaram na carga viral de PCV2 de suínos naturalmente infectados.
  • A double-antibody sandwich ELISA based on the porcine circovirus type 2 (PCV2) propagated in cell culture for antibody detection

    Few studies have described enzyme-linked immunosorbent assays (ELISAs) for the detection of antibodies against porcine circovirus type 2 (PCV2) based on antigens produced in cell culture. Furthermore, few articles have described viral purification techniques for members of the family Circoviridae. This occurs because circoviruses are difficult to isolate, noncytopathogenic, and produce low viral titres in cell culture. Thus, for overcoming these difficulties in the cultivation of PCV2, this study aimed to develop a double-antibody sandwich ELISA based on the cell culture antigen PCV2b for the quantification of anti-PCV2 antibodies. A 20% and 50% discontinuous sucrose cushion was used for viral purification, which enabled the separation of cell culture proteins in the 20% sucrose cushion and a greater viral concentration in the 50% sucrose cushion. Following isopycnic centrifugation, PCV2 was concentrated in the band with density values from 1.330 to 1.395g/cm3. Viral purification was assessed using SDS-PAGE, indirect ELISA and electron microscopy. The standardised ELISA revealed a strong linear correlation (r= 0.826, p<0.001) when compared with a commercial ELISA kit. The assay exhibited low variability (inter-assay coefficient of variation of 4.24% and intra-assay of 1.80%) and excellent analytical specificity conferred by the capture antibody produced in rabbit. Thus, this ELISA is a rapid, specific and convenient method for the detection of antibodies against PCV2 in studies of experimental and natural infection, and in monitoring the response to vaccination on commercial farms.

    <p class="Pa7" style="margin-right: 28pt;"><span style="font-family: Verdana; color: black; font-size: 8pt;">O Granuloma lepróide canino (GLC) ou “lepra canina” consiste em micobacteriose, com quadro dermatopático que conta com aspectos ainda obscuros, principalmente quanto ao seu modo de transmissão e agente etiológico envolvido. Com a finalidade de se verificar a caracterização gênica da micobactéria em questão, procedeu-se estudo a partir de cortes histológicos, de animais com diagnóstico estabelecido da enfermidade. A amostragem foi composta de 13 cães, atendidos no Serviço de Dermatologia do HOVET/USP. Tal material foi posteriormente submetido às técnicas de reação em cadeia de polimerase (PCR), “nested” e em tempo real. O método da PCR “nested” confirmou a presença do agente em apenas 18,7% dos animais. Enquanto que, em 69,2% das amostras, o DNA da micobactéria envolvida foi detectado pela PCR em tempo real, demonstrando ser neste estudo, o método de maior sensibilidade. Pôde-se concluir ainda que a espécie de micobactéria causadora do Granuloma lepróide canino nos cães brasileiros possui homologia gênica de 100%, quando comparada às amostras provindas de animais de estudos estrangeiros. <strong></strong></span></p>
