This paper outlines the underlying principles of PAT, its application in pharmaceutical manufacturing processes, and the impact on assurance of quality and reduction of cost. Real-time monitoring, multivariate data analysis, and process control strategies are three modules that are computed and integrated with PAT to develop robust and efficient manufacturing processes. A number of case studies and examples have been used to illustrate this relationship between the implementation of PAT and a reduction in variability with an improvement in process control and consistency of the product, which finally realizes million-dollar savings. It also debates the regulatory perspectives and challenges involved in PAT adoption, focusing on how stakeholders in the industry and agencies can integrate in developing and implementing innovations that will pass the test of compliance criteria. In general, what this paper presents is that PAT will drive pharmaceutical manufacturing into advancement for higher standards of quality with increased efficiency.
Fundamentals of Stem Cell Therapy in Disease Prevention Rajarshi Nath, Arijit Das, Shambo Panda, Souvik Biswas, Biplab Debnath Stem Cell Therapy in Dermatological Disorders, 2025 Stem cell therapy is a possible method of treating most diseases and restoring damaged tissue. In this review, the history, mechanism, application, problems, and prospects of stem cell therapy are summarized. Self-renewal and differentiation into other cell types are key features of stem cells. There, they can be obtained from embryonic, adult, and induced pluripotent sources. Stem cells can replace damaged cells, modulate the immune system, and secrete factors that facilitate the healing of tissues. Stem cell therapy has been researched for the treatment of neurological diseases, cardiovascular diseases, autoimmune diseases, and other conditions. However, other problems have to be solved, such as tumorigenesis risk. Recent progress in technologies of induced pluripotent stem cells and gene-editing technologies, such as CRISPR-Cas9, has provided new opportunities for personalized and targeted stem cell therapies. In the future, we also need to develop safer, more effective, and more accessible stem cell–derived therapies. Consequently, laws should be created to guarantee the clinical implementation and safe evolution of these medications. Although stem cell treatment presents great possibilities for revolutionizing regenerative medicine and enhancing patient outcomes, its advantages can only be fully appreciated by coordinated efforts among scientists, doctors, and legislators.
Simultaneous Estimation of Cal DL-2-Hydroxy,4-methyl thio-butyrate and Ca-2-oxo3-phenyl Propionate in Alpha-Ketoanalogues Tablets by Validated RP-HPLC Method Shambo Panda, Anindya Bose, Biplab Debnath, Rajarshi Nath, Arijit Das, Somsubhra Ghosh Indian Journal of Pharmaceutical Education and Research, 2024 Introduction: Alpha-Ketoanalogues is a dietary supplement drug containing Calcium-DL- 2-hydroxy-4-(methylthio) butyrate (CHMTB) and Calcium-2-oxo-3-phenyl propionate (COPP) for the management of chronic renal disease. Objectives: In this study, a simple RP-HPLC method has been developed to simultaneously determine Calcium-DL-2-hydroxy-4-(methylthio) butyrate (CHMTB) and Calcium-2-oxo-3-phenyl propionate (COPP) in Alpha-Ketoanalogues Tablets. Materials and Methods: The compounds were separated in a single run using a Waters XBridge 250 mm x 4.6 mm C18 column with 5μm particle size and 130 Å pore size. Elution was carried out via gradient mode with the UV detector wavelength set at 210 nm. The mobile phase selected was a binary mixture of ammonium hydrogen sulphate buffer (pH 7.0) and acetonitrile at a 1.0 mL/min flow rate. Results: The retention time of CHMB and COPP was found to be about 7 min and 20 min, respectively. The chromatographic method shows detector linearity in 50-150% of the operating range with the square of correlation coefficient at 0.9998 and 0.9999 for these two compounds, respectively. The parentage recovery of both CHMB and COPP was within 98-102% range. The method validation was carried out per the USP (chapter 1225) and ICH guidelines (Q2-R2). The developed method was found to be specific, precise, linear, robust, and accurate. Conclusion: This method could be utilized to assay tablet formulation of CHMB and COPP during the in-process and finished product quality control.

Olanzapine induced bruxism Tathagata Roy Journal of Medical Pharmaceutical and Allied Sciences, 2021 Iatrogenic or drug induced disease is of great clinical significance in therapeutics. Treatment with antidepressants like selective serotonin reuptake inhibitors (SSRIs) and antipsychotics reveal Bruxism (excessive grinding and clenching of the teeth) as such type of iatrogenic disease, however it is rarely found with atypical antipsychotic drugs like Olanzapine. A 50-year male patient, suffering from schizophrenia was under regular antipsychotic medication receiving Tab. Olanzapine 5 mg O.D., Tab. Clonazepam 0.5 mg O.D. regularly for past 25 years. Recently he gradually developed excessive grinding and clenching of teeth despite of any dental and ENT clinical complications. Biochemical and hematological tests also showed normal result. On admission with the complication of bruxism, tab. Olanzapine dose was reduced to 2.5 mg OD, however even after dose reduction there was not much improvement in bruxism. On the 4th day of the admission tab. Olanzapine has been substituted with tab. Clozapine 50 mg TDS. After 1 week of withdrawal of Olanzapine there was no complain of bruxism and he was discharged. On follow up after 1month the complication was occasional. Causality assessment revealed a probable relation between Olanzapine and Bruxism. Though among the atypical neuroleptics, extrapyramidal side effects are thought to be less common with olanzapine, but it may cause fatal consequences. Therefore, this case study and proper investigation may be helpful in designing the treatment strategy in more effective way and also aware all the healthcare professionals to monitor the patients on neuroleptics.

Diabetes was induced in albino Wistar rats of either sex by intraperitoneal (60mg/kg b.w.) of STZ, freshly dissolved in citrate buffer (0.01 M, pH 4.5). Ficus carica leave extract in different solution (viz. petroleum ether, ethyloacetate, methanol and aqueous) were administered to diabetic rats for 9 days. The effect of extracts on blood glucose and body weight was studies on day 1st and 9th. The study showed that the ethyl acetate, methanolic and aqueous extract of Ficus sarmentosa leaves reduced blood glucose level and body weight significantly. This may justify the use of ficus species as ethanomedical medicine for treatment of diabetes mellitus.
