Summary Brazilian Caatinga is one of the most diverse, seasonally dry, tropical forests of the world and is threatened by strong land use pressure and poor protection. One way of understanding changes in the soil conditions caused by anthropogenic activities is using the nematode assemblages as potential bioindicators. This study analysed changes in nematode communities in three different types of land use (natural forest-CA, natural area with accumulation of salts-SS, and agricultural growing areas, such as cover crops-CC for cattle and fruit trees-IC) in the Caatinga, in the sub-median region of the São Francisco River Valley (SFRV), Pernambuco, northeastern Brazil. We recorded a total of 63 262 nematode individuals, representing 42 genera, belonging to 23 families. Nematode abundance, diversity and maturity indices were highest in the CC and lowest in the SS. Among the nematode trophic groups, bacterivores, omnivores + predators and plant parasites were significantly affected by type of land use. Different soil properties were related to the differences in taxonomic composition among the natural and agricultural areas, accounting for 26.6% of the total variation. In general, our results indicate that soil salinisation and desertification in the natural areas, as well as soils subjected to cover crops and fruit tree production, significantly influenced the composition of nematode communities in the São Francisco River Valley. - Selection strategy of a Coffea canephora rootstock with simultaneous nematode resistance to Meloidogyne exigua, M. incognita and M. paranaensis

To investigate the degree of parasitism of two populations of Meloidogyne exigua, the gall index (GI) and the reproduction factor (RF) of M. exigua races 1 (Est E2) and 2 (Est E1) were analyzed in 47 progenies on F3:4 or F4:5 generation derived from the crossing between Coffea arabica cv. Catuaí Amarelo and Timor Hybrid. C. canephora cv. Apoatã IAC 2258 and C. arabica cv. Catuaí Vermelho IAC 144 were used as resistance and susceptibility checks, respectively. The genotypes that were classified as resistant or susceptible by RF were similarly classified by GI, showing a close relationship between both methodologies. The data also indicated no differences in virulence between the nematode populations, since the progenies showed similar resistance reactions to the M. exigua races 1 and 2. According to GI from the 47 mother plants evaluated, 27 progenies (57.4%) were classified as resistant to M. exigua races 1 and 2, with GI ranging from 0.0 to 1.4 and 20 progenies (42.6%) were susceptible with GI from 2.6 to 4.4. These results showed that most of the evaluated germplasm was very promising in relation to the development of new Arabica coffee cultivars with resistance to M. exigua. - A global database of soil nematode abundance and functional group composition

As the most abundant animals on earth, nematodes are a dominant component of the soil community. They play critical roles in regulating biogeochemical cycles and vegetation dynamics within and across landscapes and are an indicator of soil biological activity. Here, we present a comprehensive global dataset of soil nematode abundance and functional group composition. This dataset includes 6,825 georeferenced soil samples from all continents and biomes. For geospatial mapping purposes these samples are aggregated into 1,933 unique 1-km pixels, each of which is linked to 73 global environmental covariate data layers. Altogether, this dataset can help to gain insight into the spatial distribution patterns of soil nematode abundance and community composition, and the environmental drivers shaping these patterns. - Soil nematode abundance and functional group composition at a global scale

This study evaluated the effects of mining practices using soil nematodes as bioindicators. Soil samples represented four different subsystems found in the CeBio Research Center, including natural/undisturbed (rainforest (RF) and savannah (SA)) and disturbed (Eucalyptusspp. (EU) and grassland (GR)) subsystems. Ecological descriptorsS(3.5-9.5),d(1.22-4.23) and MI (1.9-3.23) significantly differentiated the subsystem GR (high impact) from the other subsystems. Additionally, poorly covered soil (i.e., subsystem GR) negatively influenced the composition of nematode assemblages, and primarily so for nematodes with high c-p values. Among soil variables, pH strongly influenced the distribution of soil nematodes, negatively affecting the abundance of all c-p groups and diversity of nematodes. On the other hand, pH also positively affected FF and PP trophic groups in the subsystem GR. The similarity of subsystem EU and natural subsystems (RF and SA) suggests that soil restoration withEucalyptusspp. appears to be an adequate management practice to promote the recovery of areas subjected to mining practices. - Meloidogyne javanica control by Pochonia chlamydosporia, Gracilibacillus dipsosauri and soil conditioner in tomato

Organic matter plays a fundamental role in the antagonistic activity of microorganisms against phytonematode populations on the soil. In this study, the compatibility between the fungus Pochonia chlamydosporia (Pc-12) and the rhizobacterium Gracilibacillus dipsosauri (MIC 14) was evaluated in vitro, as well as the effect of the fungus at the concentration of 5,000 chlamydospores per gram of soil, rhizobacterium at 4.65 x 10(9) cells/g of soil, and the soil conditioner Ribumin® at 10 g/pot, either alone or in combination, against Meloidogyne javanica population in tomato plants (3,000 eggs/pot). A suspension of water or Ribumin® alone was applied on the soil as negative control, while a suspension of nematode eggs was applied as positive control. The reduction in the number of galls in roots per plant was 48 and 41% for the treatments Ribumin + MIC 14 + Pc-12 and MIC 14 + Pc-12, respectively. Regarding to the number of eggs per plant, MIC 14 and Pc-12 + Ribumin led to a reduction by 26 and 21%, respectively, compared to the control treatment. Interaction between the nematophagous fungus and the rhizobacterium was positive for the nematode control, even though G. dipsosauri inhibited P. chlamydosporia growth by up to 30% in in vitro tests. - Changes in soil and nematode community after sugarcane harvest and vinasse application

Clamidósporos são estruturas de sobrevivência do fungo nematófago Pochonia chlamydosporia. Objetivou-se, com este trabalho, avaliar diferentes substratos, teores de água e formas de inóculo para a produção in vitro de clamidósporos de P. chlamydosporia. Inicialmente, testaram-se os substratos grãos de milho triturado, grãos de arroz e casca de café e os tipos de inóculo meio líquido concentrado ou diluído (1:40) e discos de cultura, colonizados por P. chlamydosporia. Posteriormente, testou-se o substrato grãos de milho triturado suplementado ou não com caldo de batata com os inóculos do fungo nas formas de discos de micélio ou suspensão aquosa. As maiores produções de clamidósporos g-1 de substrato foram obtidas nos substratos grãos de milho triturado e casca de café, e as melhores formas de inóculo foram meio líquido diluído (1:40) e discos de micélio. A suplementação do substrato grãos de milho triturado com caldo de batata não aumentou a produção de clamidósporos do fungo e a melhor forma de inóculo do fungo foi a de discos de micélio. Além disso, testaram-se os substratos canjica, grãos de arroz, casca de café e fibra de coco, umedecidos com diferentes quantidades de água; o substrato casca de café umedecido por diferentes períodos; a casca de café enriquecida ou não com sacarose e farinha de arroz e o substrato grãos de arroz esterilizado no forno micro-ondas com diferentes quantidades de água. Apenas o substrato grãos de arroz, em todas as quantidades de água testadas, apresentou maior produção de clamidósporos. Não houve diferença na produção de clamidósporos no substrato casca de café umedecido por diferentes períodos e apenas quando a casca de café foi enriquecida com farinha de arroz, apresentou maior média de clamidósporos g-1 de substrato. Todos os tratamentos apresentaram grande produção de clamidósporos g-1 quando o substrato utilizado foi o de grãos de arroz tratados no forno de micro-ondas. O melhor meio de cultivo de P. chlamydosporia para a produção de clamidósporos foi o substrato contendo grãos de arroz.
