    Parkinson’s disease (PD) affects about 8.5 million individuals worldwide. Oxidative and inflammatory cascades are implicated in the neurological sequels, that are mostly unresolved in PD treatments. However, proper nutrition offers one of the most effective and least costly ways to decrease the burden of many diseases and their associated risk factors. Moreover, prevention may be the best response to the progressive nature of PD, thus, the therapeutic novelty of honey and levodopa may be prospective. This study aimed to investigate the neuroprotective role of honey and levodopa against 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced oxidative stress. Fifty-four adult male Swiss mice were divided into control and PD model groups of 27 mice. Each third of the control mice either received phosphate buffered saline, honey, or levodopa for 21 days. However, each third of the PD models was either pretreated with honey and levodopa or not pretreated. Behavioral studies and euthanasia were conducted 2 and 8 days after MPTP administration respectively. The result showed that there were significantly (P<0.05) higher motor activities in the PD models pretreated with the honey as well as levodopa. furthermore, the pretreatments protected the midbrain against the chromatolysis and astrogliosis induced by MPTP. The expression of antioxidant markers (glutathione [GSH] and nuclear factor erythroid 2-related factor 2 [Nrf2]) was also significantly upregulated in the pretreated PD models. It is thus concluded that honey and levodopa comparably protected the substantia nigra pars compacta neurons against oxidative stress by modulating the Nrf2 signaling molecule thereby increasing GSH level to prevent MPTP-induced oxidative stress.


  • Morinda lucida and Annona muricata reduced hepatic lipid peroxidation and promoted melatonin/TNFα/p53-mediated apoptosis in sodium arsenite-induced toxicity in rats

    Arsenic-induced carcinogenesis can result in cancers of the liver in exposed organisms. This study evaluated anticancer potentials of MLF1 and AMF1 extracted from Morinda lucida and Annona muicata leaves respectively in Sodium arsenite (SA)-induced toxicity in rats. Sixty adult female rats were randomly divided into 12 groups (n = 5). Group 1 was control. Group 2 received 5-weeks administrations of 10 mg/kg bodyweight of SA. Groups 3-6 received SA-dose for 2 weeks followed by 3-weeks posttreatments with MLF1-doses and AMF1-doses respectively. Groups 7-10 received only 5-weeks administrations of MLF1-doses and AMF1- doses respectively. Groups 11 and 12 received 5-weeks co-administrations of SA-dose with highdoses of MLF1 and AMF1 respectively. Drugs/ extracts were administered orally. Liver histopathology (Heamatoxylin and Eosin) and ELISA concentrations of sera Melatonin and TNF-alpha were evaluated. Malondialdehyde (thiobarbituricacid assay) and p53 (ELISA) levels were evaluated in liver homogenates. Data were statistically analysed. Results showed normal liver histology in Groups 1-12. Post-treatments of SA-induced toxicity with MLF1 and AMF1 resulted in significant (P≤0.05) and non-significant decreased levels (P≥0.05) of Malondialdehyde, TNF-alpha and p53, but significant (P≤0.05) and non-significant increased Melatonin levels (P≥0.05) in Groups 3-12 compared with Group 2. MLF1 and AMF1 possess anticancer, antioxidant, pro-Melatonin, anti-inflammatory and hepato-protective potentials.

  • Low dose bitter leaf improves sperm quality disrupted in immunosuppressed wistar rats: An experimental study

    Background: Synthetic prednisolone (PRED) is a widely used over-the-counter glucocorticoid. Glucocorticoids have inhibitory effects on the immune system and are often used as immunosuppressive agents. Suppressed immunity may impact fertility via the hypothalamic-pituitary-adrenal axis. Bitter leaf (BL) has been shown to improve sperm parameters, but its effects on immunosuppression-associated infertility have not yet been documented.&#x0D; Objective: To determine the fertility effects of bitter leaf on immunosuppressed Wistar rats.&#x0D; Materials and Methods: A total of 30 male adult Wistar rats were randomly assigned to 6 groups (n = 5/each). Group A served as a control and were given distilled water in addition to normal feeds, group B received 2 mg/kg PRED for 14 days and served as the standard immunosuppressed group, and groups C-F were immunosuppressed as in B but in addition received 50 mg/kg levamisole, low-dose (250 mg/kg) BL, highdose (375 mg/kg) BL, and low-dose BL + levamisole, respectively. The CD4 counts, hematological parameters, and sperm parameters were analyzed and compared.&#x0D; Results: There were significant decreases in sperm motility, progressive motility, morphology, and life/death ratio in the animals given PRED only compared to the controls (p = 0.002, 0.001, 0.001, and 0.01, respectively). These were significantly increased in the treated groups, and animals given levamisole and 250 mg/kg BL showed significantly increased sperm counts compared to the controls (p = 0.04 and p = 0.04, respectively).&#x0D; Conclusion: Low-dose BL (250 mg/kg) restored the sperm parameters altered by prednisolone administration.&#x0D; Key words: Bitter leaf, Immunosuppression, Infertility, Prednisolone, Rats.

  • Prenatal exposure to gestational nicotine before neurulation is detrimental to neurodevelopment of wistar rats’ offspring

    Background and aim This study aimed to determine the effect of gestational nicotine exposure before neurodevelopment on the morphology and histology of the prefrontal cortex (PFC) in rats. Methodology Adult female Wistar rats were time-mated and grouped into three categories: (a) control–given 0.1 mL of normal saline, (b) low-dose nicotine–given 6.88 mg/ kg/d/0.05 mL, and (c) high-dose nicotine–given 13.76 mg/kg/d/0.1 mL in two divided doses. Treatment was given intraperitoneally from gestational days 2 to 6. On postnatal day 15 (P15), the pups were separated from their mothers, anaesthetised and sacrificed, followed by intracardial perfusion with 4% paraformaldehyde. PFC was excised from the brain and processed for tissue histology, histochemistry, and morphology of brain cells. Results Gestational nicotine exposure during the first week of gestation in rats significantly reduced birth weights in nicotine-treated groups compared with control; it, however, accelerated body weights, altered neuronal morphology, and elevated astrocytic count significantly, while oligodendroglial count was slightly increased in the PFC of juvenile rats examined at P15. Conclusion These alterations revealed that gestational nicotine exposure before the commencement of the cellular processes involved in brain development negatively affects neurodevelopment, and this could result in neurological dysfunctions in later life.

  • Histoenzymic effects of cigarette smoke on the superior colliculus of rattus norvegicus

    The study was conducted using twenty adult Wistar rats that were randomly grouped into a treatment and a control group comprising both sexes. The control group was exposed to smoke from cotton wool, while the treatment group inhaled smoke from a completely burnt cigarette containing 0.735 g tobacco, once daily for 14 days. The rats were sacrificed by cervical dislocation and the superior colliculi (SC) excised, weighed, homogenized and assayed for the activities of lactate dehydrogenase (LDH), glucose-6-phosphate dehydrogenase (G6PDH), and alkaline phosphatase (ALP). SC tissues for histological studies were fixed in formol calcium and subsequently processed using Feulgen reaction for nucleic acids and Cresyl violet stain for Nissl bodies studies. There was an increase in the enzyme activities in the treated group, compared to the control; and this increase was more in the female rats although not statistically significant. The derangements observed in the enzyme studies led to various histoarchitectural changes in the SC of the rats exposed to tobacco smoke, and a gradual depletion of DNA positive cells and Nissl bodies in the neurons.
