Chlorpyrifos is an organophosphate class pesticide to which humans are constantly exposed. Currently, there are few studies that evaluate the effects of this insecticide on the epididymis; therefore, the objective of the present study was to assess the effects of chlorpyrifos on the postnatal development of the epididymis during the peripubertal period. Thirty Wistar rats were divided into three groups: Control, C5, and C15. The control group received only the vehicle and the C5 and C15 groups received, respectively, 5 and 15 mg/kg of chlorpyrifos via gavage during 40 days (DPN25–DPN65). On DPN 66, the animals were euthanized. The epididymis was used for sperm count, histopathological, stereological, and oxidative stress analyses. Sperm from the vas deferens was used for motility and mitochondrial activity analyses. There was a difference in the proportion of luminal, stromal, and epithelial compartments in the groups exposed to chlorpyrifos. An increase in sperm transit time in the caput epididymis was also observed in the C5 group and a decrease in mitochondrial activity in sperm from the C15 group. Oxidative stress analyses showed a decrease in GST activity in the C5 and C15 groups. Sperm motility remained unchanged. These results suggest that the exposure to chlorpyrifos during peripuberty harms the postnatal development of the epididymis. This study is the first to evaluate the morphology and function of the epididymis in peripubertal rats.

The present study aims to evaluate the effects of LDX administration on the hepatic morphophysiology of pubertal Wistar rats, since the liver plays a central role in systemic metabolism. Male Wistar rats were randomly distributed into two experimental groups: LDX group (LDX) — received 11.3 mg/kg/day of LDX diluted in tap water; and the Control group (C) — received tap water only. Animals were treated by gavage during puberty, from postnatal day (PND) 25 to PND 65. The results showed that administration of LDX decreased white adipose tissue weight without altering overall body weight gain. Plasma biochemical analysis demonstrated an increase in plasma total cholesterol and in the concentration of hepatic transaminases in LDX rats. In addition, LDX animals exhibited an increase in N-acetyl-β-D-glucosaminidase activity, indirectly indicating macrophage recruitment to the liver. This study shows that daily exposure to LDX during puberty causes early liver inflammation and damage due, in part, to increased hepatic transaminases and associated with indirect signs of macrophage recruitment. This finding highlights the importance of monitoring the indiscriminate use of amphetamines and alerting patients to possible liver disorders. Graphical Abstract

Thus far, no reports of its presence or connection to pathological processes have been described in the peripheral nervous system. Our study demonstrates the presence of a PNN in the spinal afferent innervation of the distal colon of mice and characterizes structural and morphological alterations induced in an ulcerative colitis (UC) model. C57Bl/6 mice were given 3% dextran sulfate sodium (DSS) to induce acute or chronic UC. L6/S1 dorsal root ganglia (DRG) were collected. PNNs were labeled using fluorescein‐conjugated Wisteria Floribunda (WFA) l lectin, and calcitonin gene‐related peptide (CGRP) immunofluorescence was used to detect DRG neurons. Most DRG cell bodies and their extensions toward peripheral nerves were found surrounded by the PNN‐like structure (WFA+), labeling neurons' cytoplasm and the pericellular surfaces. The amount of WFA+ neuronal cell bodies was increased in both acute and chronic UC, and the PNN‐like structure around cell bodies was thicker in UC groups. In conclusion, a PNN‐like structure around DRG neuronal cell bodies was described and found modulated by UC, as changes in quantity, morphology, and expression profile of the PNN were detected, suggesting a potential role in sensory neuron peripheral sensitization, possibly modulating the pain profile of ulcerative colitis. image

There are still no data in the literature relating the possible toxic effects of LDX in the kidney. Therefore, the present study aims to evaluate the effects of LDX exposure on morphological, oxidative stress, cell death and inflammation parameters in the kidneys of male pubertal Wistar rats, since the kidneys are organs related to the excretion of most drugs. For this, twenty male Wistar rats were distributed randomly into two experimental groups: LDX group—received 11,3 mg/kg/day of LDX; and Control group—received tap water. Animals were treated by gavage from postnatal day (PND) 25 to 65. At PND 66, plasma was collected to the biochemical dosage, and the kidneys were collected for determinations of the inflammatory profile, oxidative status, cell death, and for histochemical, and morphometric analyses. Our results show that there was an increase in the number of cells marked for cell death, and a reduction of proximal and distal convoluted tubules mean diameter in the group that received LDX. In addition, our results also showed an increase in MPO and NAG activity, indicating an inflammatory response. The oxidative status showed that the antioxidant system is working undisrupted and avoiding oxidative stress. Therefore, LDX‐exposition in male rats during the peripubertal period causes renal changes in pubertal age involving inflammatory mechanisms, antioxidant activity and apoptosis process.

This study reports the development of a carbomer hydrogel containing biogenic silver nanoparticles (bioAgNPs) and its effectiveness in wound treatment. This hydrogel showed in vitro bactericidal activity after 2 h, according to the time–kill assay. It also reduced bacterial contamination in rat wounds without impairing their healing since the hydrogel hydrophilic groups provided hydration for the injured skin. The high number of inflammatory cells in the first days of the skin lesion and the greater degree of neovascularization one week after wound onset showed that the healing process occurred normally. Furthermore, the hydrogel-containing bioAgNPs did not cause toxic silver accumulation in the organs and blood of the rats. This study developed a bioAgNP hydrogel for the treatment of wounds; it has a potent antimicrobial action without interfering with cicatrization or causing silver bioaccumulation. This formulation is effective against bacteria that commonly cause wound infections, such as Pseudomonas aeruginosa and Staphylococcus aureus, and for which new antimicrobials are urgently needed, according to the World Health Organization’s warning.
