  • Activity of Stingless Bee Honey (Heterotrigona itama) Hydrogel Against Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa

    Diabetic foot ulcers (DFUs), a prevalent and debilitating complication of diabetes mellitus (DM), are commonly exacerbated by polymicrobial infections involving pathogens. Recent investigations have highlighted the therapeutic potential of stingless bee honey (SBH) derived from Heterotrigona itama, which exhibits significant antibacterial activity attributed to its bioactive constituents. This study aimed to evaluate and compare the antibacterial efficacy of 20% and 25% SBH-based hydrogels against Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa by analyzing inhibition zone diameters. A posttest-only controlled experimental design was employed. The test groups consisted of 20% and 25% SBH hydrogels, Cutimed® gel as the positive control, and sterile distilled water as the negative control. Antibacterial activity was statistically evaluated using one-way analysis of variance (ANOVA), with subsequent application of Duncan's post hoc test to determine significant differences among experimental groups. Significant differences in inhibition zones were observed among treatments (p < 0.001). The 25% honey hydrogel demonstrated the largest mean inhibition zones against all tested bacterial strains, surpassing those of the 20% formulation, the negative control, and Cutimed® gel. SBH hydrogel exhibits concentration-dependent antibacterial activity, with the 25% concentration displaying superior antibacterial effects. These findings underscore its potential as a natural therapeutic agent for managing diabetic foot ulcers, warranting further clinical investigation.
  • Antibacterial and Wound Healing Activity of Ethanolic Extract Melastoma malabathricum L

    Melastoma malabathricum L are plants that are widely found in South Kalimantan. This plant grows along the riverbanks. Flowers of M. malabathricum L have antibacterial activity greater than leaves in Staphylococcus aureus bacteria. Every fase of M. malabathricum L flowers contents different quercetin and kaempferol. M. malabathricum L flower phase consists of bud phase 1 (K1), bud phase 2 (K2), bud phase 3 (K3) and blooming phase (M). Quercetin and kaempferol are also found in M. malabathricum L fruit (F). The highest content of quercetin and kaempferol is in the blooming phase. Quercetin and kaempferol have antibacterial and wound healing activity. This research aims to analyze the antibacterial and wound healing activity of ethanolic extracts of M. malabathricum L flower (FL) and fruit (FR). The method antibacterial activity is used the liquid dilution method and continued with planting on agar media. The wound healing activity was tested on rat that were smeared with cream for 14 days and examined the histopathology of the skin. Concentration of extract is 1%, 2% dan 4%. The results showed that the skin smeared with FLC (flower cream) 4% had formed adnesia, while the other treatments had not yet formed.
  • Antioxidant and cytotoxicity activities of karamunting (Melastoma malabathricum L.) fruit ethanolic extract and quercetin

    Melastoma malabathricum L. is a type of plant naturally grows in Kalimantan that has medicinal properties. Ethanolic extract of M. malabathricum L. flower has quercetin and kaempferol contents that have antioxidant and anticancer activities. But the antioxidant and cytotoxicity activities of M. malabathricum L. fruit ethanolic extract were not known. This study measured the quercetin and kaempferol level in M. malabathricum L. fruit ethanolic extract using HPLC MS/MS, antioxidant activity using DPPH method, and cytotoxicity activity using Brine Shrimp Lethality Test (BSLT) method. Results showed the level of quercetin and kaempferol from M. malabathricum L fruit ethanolic extract are 67.78 µg/g and 43.52 µg/g, respectively. Beside, the antioxidant activity with by IC50 was 16.82±0.24 ppm and 7.38±0.41 ppm. The cytotoxicity activities of M. malabathricum L. fruit ethanolic extract and quercetin are shown by the LC50 of 313.44 ppm (95%CI 283.97-344.43) and 37.24 ppm, respectively.
  • Analysis of quercetin and kaempferol levels in various phase of flowers melastoma malabathricum L.

    Melastoma malabathricum L. flowers are part of a plant that contains quercetin and kaempferol. Quercetin and kaempferol are useful for their antioxidant and anticancer properties. The flowering phase affects levels of quercetin and kaempferol. There are four phases of flower blooming: 1st bud phase (K1), 2nd bud phase (K2), 3rd bud phase (K3), and flower bloom (M). It remains to be determined which phase of Melastoma malabathricum L. flowering shows the highest levels of quercetin and kaempferol. Quercetin and kaempferol levels were measured using HPLC MS/MS. The measured quercetin levels of K1, K2, K3, and M phases, respectively, were: 19.47 μg/g, 17.78 μg/g, 31.2 μg/g, and 94.32 μg/g. Levels of kaempferol during K1, K2, K3, and M, respectively, were: 140.99 μg/g, 57.28 μg/g, 95.32 μg/g, and 349.37 μg/g.
  • Oxidants-antioxidants profile in the breast cancer cell line MCF-7

    Reactive oxygen species (ROS) have various biological effects and they are non-linear in characteristic. In high oxidative stress, they may cause cytotoxicity, inhibit cell proliferation, and induce cell death in the form of apoptosis/necrosis; while in low or medium oxidative stress, ROS may cause DNA damage, cell mutation, inflammation, cell proliferation, and eventually they may induce carcinogenesis. Antioxidants are compounds with the ability to reduce ROS. Cell line MCF-7 is one of the breast cancer cell lines that is known to have small amount of antioxidant MnSOD compared to the other cell lines. Low antioxidant MnSOD level in breast cancer cell line MCF-7 leads to low concentration of hydrogen peroxide, because antioxidant MnSOD will convert radical superoxide to hydrogen peroxide. The aim of this research was to analyze oxidants and antioxidants profile in breast cancer cell line MCF-7 and their relationship with cell number. Observations were conducted for 5 days. The cell number was counted with tryphan blue method and haematometer. The concentration of radical superoxide was measured with DHE staining using LSCM tipe Olympus Fluoview FV 1000-Ver 1.7. MnSOD activity, hydrogen peroxide concentration, and catalase activity were measured with ELISA. The results showed that the longer of observation, the greater concentration of oxidants and MnSOD activity, but there was no change in catalase activity. Conclusion the increase in cancer cells number is influenced by radical superoxide.
