PRODUCTIVE AND MARKET ANALYSIS OF BETA-CAROTENE OBTAINED FROM THE MICROALGA DUNALIELLA SP.: CURRENT SCENARIOS AND FUTURE PERSPECTIVES

Objetivo: Apresentar o mercado de carotenoides com foco no betacaroteno extraído de Dunaliella sp., microalga capaz de sintetizar quantidades expressivas de betacaroteno.Referencial teórico: O betacaroteno é um pigmento natural com aplicabilidade na indústria alimentícia, sendo precursor de vitamina A e atuando na prevenção de diversas doenças. As microalgas do gênero Dunaliella sp. poduzem maiores quantidades de betacaroteno em relação a outras fontes naturais, porém, seu custo de cultivo limita sua competitividade.Método: Utilizou-se uma abordagem quali-quantitativa para sintetizar dados de literatura científica, relatórios e bases de dados mercadológicas. Realizou-se uma busca patentométrica na base Derwent Innovation Index (DII), onde foram identificadas 213 citações de patentes.Resultados e conclusões: O mercado global de betacaroteno é crescente, porém o Brasil possui baixa participação e dependente de importação. A demanda por Dunaliella sp. para obtenção de betacaroteno também cresce, contudo seu custo produtivo não é competitivo. As patentes indicam interesse em otimizar os meios de cultivo e de melhorar geneticamente as cepas de microalgas para viabilizar a produção.Implicações da pesquisa: O estudo busca contribuir com o melhor entendimento sobre o potencial do betacaroteno obtido de Dunaliella sp.Originalidade/valor: A produção de biomassa de Dunaliella sp. apresenta vantagens ambientais em relação ao cultivo de culturas terrestres, como a não utilização de terras agricultáveis para produção de alimentos juntamente com a possibilidade biorremediação de efluentes como processo ambientalmente amigável de cultivo, alinhando-se a diversos ODS-ONU.

The brown macroalgae Dictyota menstrualis constitutes an organism with a rich and diverse metabolism in terms of the chemistry of natural products. It is known to mainly produce diterpenes, in addition to sterols, vitamins and polysaccharides with antiviral, anti-inflammatory, anticoagulant and antioxidant properties. Despite its vast metabolic arsenal, most studies are directed to the identification of already known diterpenes, opening the field of investigation for new approaches and discoveries. Knowing this, this work aims to chemically characterize the macroalgae D. menstrualis through the technique of Ultra Efficiency Liquid Chromatography associated with High-Resolution Mass Spectrometry (UPLC-HRMS) and perform a biological screening of potential antioxidants and antifungals through bioautography. For this, specimens of D. menstrualis were collected in the São Pedro São Paulo Archipelago (PE, Brazil) and attended to ingestion with dichloromethane and fractionation by column chromatography with silica gel. The extract and fractions were then analyzed by UPLC-HRMS with ElectroSpray Ionization (ESI) in positive mode. In addition, bioautography assays were performed to evaluate the antioxidant potential with DPPH and the antifungal potential against the Cladosporium sphaerospermum . The UPLC-HRMS analyzes allowed identifying diterpenes, sterols, faty acids, glycerolipids, derivatives of photosynthetic pigments and carotenoids. Bioautography assays suggested antifungal and antioxidant potential of the organic extract and fractions. Thus, the results suggest that D. menstrualis has pharmacological potential to be explored and that studies on its chemical composition using modern techniques such as UPLC-HRMS can lead to the discovery of new structures and reinforcement of the knowledge of the chemistry of this algae.

ABSTRACT Heavy metals (HMs) are natural components of the Earth’s crust that might originate from natural and anthropogenic sources. In excess quantities, the presence of these metals is harmful for both environment and human health. Taking this into account, various investigators examined bioaccumulator species in order to reduce environmental toxicity, among these Baccharis trimera. Therefore, the present study aimed to determine the capacity of B. trimera to bioaccumulate HMs and assess consequent cytogenotoxicity following exposure. B. trimera vegetative parts were collected from two groups (1) control, in which plants were cultivated in soil exposed to distilled water, and (2) exposed, in which plants were cultivated in soil exposed to HMs including manganese (Mn), iron (Fe), lead (Pb), copper (Cu), cobalt (Co), zinc (Zn), and chromium (Cr). HMs were quantified in cultivation soil and extracts (aqueous and ethanolic) as well as infusion of B. trimera vegetative parts. Root lengths and cytogenotoxic effects were determined using Allium cepa test. Results demonstrated that all HMs studied were absorbed and bioaccumulated by B. trimera. Root lengths were decreased when exposed to ethanolic extract of B. trimera cultivated in soil exposed to HMs solution, which was the extract that exhibited the highest cytogenotoxicity values. Thus, data demonstrated that B. trimera might serve as a bioaccumulator for the reduction of environmental toxicity associated with the presence of certain HMs.

ABSTRACT Agroecology, the application of ecological concepts to agricultural production, has been developing over the last years with consequent promotion for discovery of bioactive compounds to control pests and abolish crop diseases. In this context, algae from Nitella genus are characterized by high potential for bioeconomic applications due to (1) available biomass for harvesting or cultivation and (2) production of allelochemicals, which present a potential to protect field crops from insect infestation. Therefore, this study aimed to determine primary and secondary metabolites derived from aqueous and hydroethanolic extracts of Nitella furcata and to evaluate phytotoxic, cytogenotoxic, insecticidal, and pro-oxidative activities of these extracts. Determination of metabolites showed the presence predominantly of carbohydrates, proteins, phenols, and flavonoids in hydroethanolic extract. Both extracts of N. furcata interfered in the germination of seeds and development of seedlings of Lactuca sativa, with hydroethanolic extract exhibiting greater inhibition. Both extracts also interfered with meristematic cells of Allium cepa as evidenced by chromosomal alterations and higher pro-oxidative activity. Aqueous extract at 5 and 0 mg/ml produced 100% insect mortality. Further, hydroethanolic extract at 0 mg/ml was lethal immediately upon exposure. Therefore, results demonstrate that N. furcata is potential algae species to be considered for development of environmental and ecotoxicological studies as a source of compounds with potential use in agroecological strategies.

Bioprocess for mass production and feed utilization of Azolla pinnata in aquaculture ponds: a perspective of bioeconomy and eco-friendly technology for small farms

Aquatic plants have a high potential to be used as eco-friendly technology in fish farming effluent treatment systems. However, there is still a reduced use of the vegetable biomass produced in these treatment systems. Thus, the aim of this study was to develop an alternative feed with the aquatic plant Azolla pinnata to take advantage of plant biomass, reusing the plant to develop a new product and promote a circular economy. A. pinnata was implemented in decantation pond that receives effluents from fish farming. Samples were collected weekly at four times (0, 7, 14, and 21 days). Posteriorly, A. pinnata was processed to obtain the meal. Azolla meal was offered along with three commercial feed (24%, 28% and 32% of the crude protein) in an experiment to evaluate the zootechnical performance of tilapia in small tanks. The results indicated that A. pinnata was efficient to retention of phosphorus and nitrogen after seven days. In the experiment with animals, the treatment using commercial feed with 28% of the crude protein + Azolla showed the best efficiency rates for using the diet. Thus, the use of the A. pinnata meal was a viable alternative in the search for sustainable products to promote a bioeconomy in the small fish farms.


Social and environmental indicators of family fish farms in the atlantic forest area in the Ribeira Valley-SP

RESUMO O objetivo deste trabalho consistiu na aplicação de indicadores socioambientais em pisciculturas familiares localizadas na região do Vale do Ribeira, em área de Mata Atlântica, no sudoeste do estado de São Paulo. Para isso, foram identificadas 84 propriedades. Dessas, 40 foram selecionadas como unidades amostrais. Entre as principais características identificadas, 32% possuem lâmina d’água inferior a cinco hectares, com viveiros escavados em sistema semi-intensivo, 58% utilizam mão de obra familiar e 23% apontam a ausência de assistência técnica especializada como o principal problema enfrentado. A tilápia-do-nilo (Oreochromis niloticus) representa 57% das espécies produzidas em sistemas de monocultivo ou policultivo. Contudo, as pisciculturas familiares são classificadas como sistemas produtivos de pequeno porte, com destaque para a necessidade de adequação de recursos naturais, da capacidade de gestão e da eficiência das práticas de produção. Os indicadores sociais variaram de 0,75 a 1,00 para equidade salarial, proporção de autoemprego, uso de mão de obra local, inclusão de gênero e inclusão etária, demonstrando a capacidade que a aquicultura tem para auxiliar no desenvolvimento social local, por meio da geração de emprego e renda.

Estimating daily global radiation from common meteorological variables in a humid subtropical environment

O conhecimento do potencial solar é considerado uma informação essencial no dimensionamento e simulação do desempenho de sistemas que convertem radiação solar em energia elétrica ou calor disponível em fluídos, bem como em estudos agrícolas voltados ao manejo de irrigação e avaliação da produtividade potencial. Nem sempre as informações sobre a radiação solar incidente estão disponíveis e, por isso, torna-se importante o desenvolvimento de métodos capazes de estimar com acurácia a radiação solar. O presente estudo avaliou o desempenho de diferentes métodos de estimativa da radiação solar global (RG) em ambiente subtropical úmido brasileiro, a fim de determinar qual deles apresenta maior acurácia. As bases de dados meteorológicos utilizadas foram registradas pela estação meteorológica automática pertencente ao Instituto Nacional de Meteorologia (INMET) e instalada em Iguape, município localizado na porção paulista do Vale do Ribeira. Modelos empíricos foram calibrados e tomados como referência na avaliação do desempenho da Máquina de Vetores de Suporte (SVM) utilizando diferentes conjuntos de variáveis de entrada. Dentre os métodos testados, a SVM utilizando o conjunto de varáveis de entrada {R0, Tmax, Tmin, es max, es min} apresentou o melhor desempenho.Palavras-chave: modelos empíricos; Vale do Ribeira; SVM. ESTIMATING DAILY GLOBAL RADIATION FROM COMMON METEOROLOGICAL VARIABLES IN A HUMID SUBTROPICAL ENVIRONMENT  ABSTRACT: Knowledge of solar potential is essential in the design and simulation of systems that convert solar radiation in electric power or available heat in fluids, as well as in agricultural studies focused on irrigation management and potential productivity assessment. Oftentimes, information about solar radiation are not available and, therefore, it becomes important the development of methods for obtain reliable data of solar radiation. The present study evaluated the performance of different methods of estimating global solar radiation (RG) in a Brazilian humid subtropical environment in order to determine which is more accurate. The meteorological databases used were registered by the automatic meteorological station belonging to the National Institute of Meteorology (INMET) and installed in Iguape, a municipality located in the São Paulo portion in the Ribeira Valley. Empirical models were calibrated and taken as reference in the performance evaluation of the Support Vector Machine (SVM) using different sets of input variables. Among the tested methods, the SVM using the set of input variables {R0, Tmax, Tmin, esmax, es min} presented the best performance.Keywords: empirical models; Ribeira Valley, SVM.


Nutritional value changes in response to temperature, microalgae mono and mixed cultures

Abstract Aim The response of mixed cultures and monocultures of Pseudopediastrum boryanum (Turpin) E. Hegewald and Scenedesmus obliquus (Turpin) Kützing was assessed in the laboratory at three different temperatures: 20, 30 and 40 °C. The change in biomass and biochemical composition of these cultures was evaluated. Methods Microalgae were placed in a batch culture over 21-days in ASM1 medium. Cell density was directly counted every two days using a Fuchs-Rosenthal hemocytometer. Chlorophyll ‘a’ and total carotenoids were extracted twice. Protein, carbohydrate, total lipid and fatty acid contents were determined at the end of the experiment. Results Cultures grown in 40 °C exhibited no growth. However, there was increased cell density (13.6 x 106 cell.mL-1) and biomass (55 g.L-1) in the S. obliquus monoculture at 30 °C. High protein concentrations (672.6 mg.g-1) were observed in monocultures P. boryanum and S. obliquus at 20 °C treatments. There were high carbohydrate concentrations (6.17 mg.g-1) in P. boryanum unialgal at 30 °C. There was no significant difference in total lipid content between S. obliquus (95.5 mg.g-1), P. boryanum (96.3 mg.g-1) and the mixed culture at 20 °C (105.3 mg.g-1). FAMEs varied significantly regarding the number of unsaturated components, which predominantly consisted of fatty acids with two or more unsaturated bonds. Conclusions The biomass of the treatments analyzed was rich in proteins and essential fatty acids (such as linolenic acid), yet low in carbohydrate content, suggesting its potential use as a food supplement. Our results indicate that mixed culture of P. boryanum and S. obliquus was inefficient in cultivating biomass or biochemical compounds as compared to the unialgal cultivations. However, algae grown together showed better stability in their biochemical composition in response to changes in temperature, an important factor for microalgae production in open ponds and in food safety measures. These results suggest the consortia among different species of microalgae should be tested to determine better methodologies for the production of biomass and metabolites with greater stability towards environmental factors.


IET for the environmental monitoring of the intensive production of tilapia in cage in the Atlantic forest region of South America

A produção de tilápias (Oreochromis niloticus) em tanques-rede exige o constante monitoramento das propriedades físico-químicas da água, pois é uma atividade considerada potencialmente poluidora do ambiente aquático e o monitoramento é fundamental para evitar processos de eutrofização do corpo hídrico. O presente estudo avaliou a influência da produção intensiva de tilápia do Nilo em tanques-rede sobre o grau de trofia e qualidade da água em uma represa localizada no Vale do Ribeira, região de Mata Atlântica no sudeste do Brasil. Na classificação do grau de trofia foram considerados cinco classes, que foram estabelecidas a partir do Índice do Estado Trófico (IET). O experimento foi conduzido em três períodos: antes (90 dias), durante (180 dias) e após (90 dias) à produção intensiva de peixes na represa. Instalaram-se 30 tanques-rede de 4 m3 com densidade de estocagem de 150 peixes por m3. A partir dos resultados experimentais, verificou-se que o IET foi crescente no período de produção passando de ultraoligotrófico à mesotrófico, alcançou valor máximo de 53,2 ± 0,4 e após a produção manteve-se oligotrófico. Os parâmetros de qualidade de água não apresentaram diferenças significativas entre os períodos analisados (p>0.05). A aplicação com base no IET representa uma ferramenta prática para o monitoramento ambiental e análise de riscos da produção de peixes no ambiente aquático.

Chemical analysis and toxicity of seaweed extracts with inhibitory activity against tropical fruit anthracnose fungi

BACKGROUND Banana and papaya are among the most important crops in the tropics, with a value amounting to millions of dollars per year. However, these fruits suffer significant losses due to anthracnose, a fungal disease. It is well known that certain seaweed extracts possess antifungal activity, but no published data appear to exist on the practical application of this property. In the present study, five organic Brazilian seaweed extracts were screened for their activity against banana and papaya anthracnose fungi. Furthermore, cytotoxic and mutagenic effects of the extracts were evaluated by the brine shrimp lethality assay and the Allium cepa root-tip mutagenicity test respectively, while their major components were identified by gas chromatography/mass spectrometry. RESULTS Strong fungus-inhibitory effects of Ochtodes secundiramea and Laurencia dendroidea extracts were observed on both papaya (100 and 98% respectively) and banana (89 and 78% respectively). This impressive activity could be associated with halogenated terpenes, the major components of both extracts. Only Hypnea musciformis extract showed cytotoxic and mutagenic effects. CONCLUSION The results of this study suggest the potential use of seaweed extracts as a source of antifungal agents with low toxicity to control anthracnose in papaya and banana during storage.


Influence of laboratory cultivation on species of Rhodophyta physiological evaluations and antifungal activity against phytopathogens

O presente estudo verificou as alteracoes ocorridas na fisiologia de tres especies de Rhodophyta por meio da analise da concentracao de pigmentos e de carboidratos nao estruturais, avaliando comparativamente algas coletadas da natureza e apos 28 dias de cultivo em laboratorio. Outro objetivo do estudo foi determinar se as especies mantem o potencial antifungico apos o periodo de cultivo em laboratorio, correlacionando as mudancas ocorridas na fisiologia com as de bioatividade. Apos a realizacao de cultivo em laboratorio os resultados indicaram a tendencia de reducao nas concentracoes de pigmentos nas tres especies de macroalgas, principalmente para as ficobiliproteinas. Para os carboidratos nao estruturais nao foi observado padrao de resposta das macroalgas em relacao as condicoes avaliadas. Os extratos das macroalgas Ochtodes secundiramea e Palisada flagellifera, apos cultivo em laboratorio, apresentaram aumento significativo do potencial antifungico positivamente correlacionado com a alteracao na concentracao e nas razoes de clorofila a e pigmentos acessorios. A aplicacao de tecnicas de cultivo em laboratorio das macroalgas foi uma ferramenta eficiente para o conhecimento da fisiologia, obtencao de biomassa e de compostos com aplicacao biotecnologica, sem impactar populacoes naturais.


Fungitoxic effect of essential oil from aroeira (Schinus terebinthifolius RADDI) on Colletotrichum gloeosporioides

Neste trabalho foi avaliado o efeito do óleo essencial do fruto de Schinus terebinthifolius sobre o crescimento micelial do fungo Colletotrichum gloeosporioides in vitro, e no desenvolvimento da antracnose no período de pós-colheita em mamões. As diferentes concentrações de óleo foram diluídas em Tween 80 a 8%. No experimento in vitro foram preparados meios de cultura BDA nas concentrações de 0,05; 0,10; 0,25 e 0,50% do óleo essencial. O controle negativo foi realizado apenas com meio BDA e o controle solvente com meio BDA e Tween 80 a 8%. A inibição do crescimento do fungo foi diretamente proporcional à quantidade do óleo e a maior inibição encontrada foi de 79,07% na concentração de óleo de 0,50%. No experimento in vivo os frutos do mamoeiro foram inoculados com o fungo em quatro tratamentos: com biofilme; com biofilme mais 0,50% do óleo; com fungicida Prochloraz e frutos controle. Embora o tratamento com óleo tenha sido eficiente contra o fungo, não foi indicado comercialmente, pois apresentou valores elevados de perda de massa fresca, de firmeza, e também sintomas de fitotoxidade. O óleo tem propriedade antifúngica contra C. gloeosporioides in vitro e in vivo, contudo, não é recomendado para o mamão em função da fitotoxidez
