Optimized production and characterization of ulvan from Ulva Lactuca with in vitro biological activities Amira M. Abu-Resha, Mostafa M. El-Sheekh, Ghada W. Abou-El-Souod, Hanaa Morsi, Mofida Makhlof Scientific Reports, 2026 Bioactive sulfated polysaccharides, including ulvan, known for their broad therapeutic uses, are abundant in green algae. In this study, Ulva lactuca ulvan was produced under optimized conditions involving temperature, pH, extraction time, and solvent concentration adjustments. The extracted ulvan ( U. lactuca ulvan, ULU) was identified using analytical methods such as Fourier Transform Infrared Spectroscopy (FTIR), Thermogravimetric Analysis (TGA), X-ray Diffraction (XRD), Scanning Electron Microscopy (SEM), High-Performance Liquid Chromatography (HPLC), and Energy-Dispersive X-ray Spectroscopy (EDX) to confirm its compositional stability. A maximum yield of 23.33 ± 0.28% was achieved, with rhamnose as the main sugar, accounting for 14.10 µg/g of dried weight. Ulvan’s biological activities are likely influenced by its sulfate content, which measured 35.35 ± 0.25%. The bioactivity of ULU was tested for anticancer, antioxidant, and antiviral effects. ULU showed significant anticancer activity against pancreatic cancer cells (Panc-1). At a dose of 1000 µg/ml, ULU achieved an inhibition rate of 86.15%, with an IC 50 of 123.51 ± 10.95 µg/ml, demonstrating substantial effectiveness in inhibiting pancreatic cancer cell growth. In antioxidant tests, ULU exhibited moderate free radical scavenging activity against DPPH, with a maximum inhibition rate of 88.31% ±2.64% at 1000 µg/ml and an IC 50 of 263.73 ± 9.41 µg/ml. These results highlight the benefits of improved extraction and optimized conditions, indicating that ULU has potential as a natural antioxidant, though with modest efficacy compared to ascorbic acid. ULU also demonstrated limited antiviral activity, with an inhibition rate of 40.25 ± 2.61% against Hepatitis A virus (HAV) at 50 µg/ml, as measured by the MTT assay. The cytotoxicity test showed a CC₅₀ value of 230.53 ± 2.09 µg/ml, indicating a moderate safety margin. Despite its limited antiviral effectiveness, further research into combining ULU with other drugs or exploring synergistic effects could enhance its therapeutic potential.
Antimicrobial, antidiabetic, antiviral, and antioxidant activities of fucoidan extracted from the brown seaweed Padina pavonica Faiza M. A. Akl, Mostafa M. El-Sheekh, Mofida E. M. Makhlof, Suzan I. Ahmed BMC Biotechnology, 2025 Macroalgae are considered a promising biological source of active substances, and sulfated polysaccharides are among the most important compounds with potential applications. Here, fucoidan from Padina pavonica collected from the Rocky Bay of Abu Qir in Alexandria was analyzed for its ash, water, protein, sulfated groups, elemental content, total sugars, and uronic acid levels. Additionally, its monosaccharides were qualitatively and quantitatively determined using high-performance liquid chromatography for fucose detection, which is the main fingerprint of fucoidans. The isolated fucoidan was characterized using FTIR, scanning electron microscopy, EDX, and thermogravimetric analysis (TGA). Its biological activities, including antiviral, antioxidant, antimicrobial, and antidiabetic effects, were then evaluated. The yields of fucoidan, fucose, and sulfate were found to be 17.8 ± 0.23%, 34.45%, and 9.52 ± 0.19%, respectively. It inhibited HSV-1 with an inhibition percentage of 30.89 ± 0.84. The maximum Ferric Reducing Antioxidant Power (FRAP) value reached 81.82 ± 1.44% at 1000 µg/ml. Padina fucoidan (PF) showed the largest inhibition zone of 18 mm against Methicillin-Resistant Staphylococcus aureus (MARSA) (ATCC 4330) with an MIC of 1.25 mg/L. It also demonstrated a promising inhibitory effect on α-Amylase enzyme, reaching 75.69 ± 1.05 at a concentration of 1000 µg/ml. We conclude that Padina pavonica is an excellent producer of fucoidan, with a significant sulfate content that enhances its biological activities, especially its antidiabetic properties.
Characterization and Phenolic Profiling of Anticandidal Polycladia myrica and Its Mediated Iron Nanoparticles with the Evaluation of Their Antioxidant, Anti-Alzheimer, Catalytic Degradation, and Anticancer Activity via the P53 Pathway Abeer I. M. EL-Sayed, Fatma Alzahraa Mokhtar, Mohammad Y. Alfaifi, Hanan Khalaf Anazi, Mofida E. M. Makhlof ACS Omega, 2025 As a potent reducing and capping agent, Polycladia myrica extract was used to create iron nanoparticles (FeNPs). UV–visible, Fourier transform infrared (FTIR), X-ray diffractive analysis (XRD), scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDX), zeta potential, hydrodynamic analysis with the determination of polydispersity index (PDI), and transmission electron microscopy (TEM) were used to characterize the produced iron nanoparticles. By examining the peak at 320 nm, UV–visible spectroscopy confirmed the production of PFeNPs. Additionally, various in vitro biological assays were conducted, demonstrating significant therapeutic potential. The cytotoxic assay was performed using lung carcinoma cells (A-549) and normal human lung fibroblast (MRC-5) cell lines, which demonstrated promising safe results with IC50 = 225.91 ± 8.93 μg/mL and CC50 = 488.80 ± 17.23 μg/mL, respectively, due to the forced apoptosis caused by the accumulation of both ROS and p53 levels inside cancer cells. PFeNPs demonstrated weak α-amylase inhibition with a percent of 33.96 ± 3.12 and potent acetylcholinesterase inhibition with a percent of 71.42 ± 3.64. The anticandidal activity of FeNPs biofabricated from P. myrica against C. albicans was estimated. The inhibition zone diameters of PFeNPs and Nystatin reference drug (mean ± SD) were about 18.82 ± 0.87 and 19.74 ± 0.98, respectively. MIC was determined to be about 500 and 312.5 μg/mL for both our algal FeNPs and the standard used drug, respectively. These results were confirmed by scanning electron microscopy, which revealed lysis and bursting of the exterior cell surface, along with deformation and death of Candida albicans cells in treated Candida isolate. These results strongly suggested the possibility of introducing PFeNPs as a cotherapy for Candida infections in diabetic cancer patients.
