  • Study on MTHFR (C677T & A1298C) Gene Polymorphisms in the Condition of Glucose Intolerance During Pregnancy

IntroductionGestational diabetes mellitus (GDM) is a prevalent medical complication in pregnancy, characterized by glucose intolerance. The global expected prevalence of GDM is approximately 15.1%. This study builds upon previous research by investigating elevated hematological parameters and exploring MTHFR gene polymorphisms in GDM.Materials and MethodsThis study included 304 pregnant women, comprising 152 patients with GDM and an equal number of normal pregnant women. Employing PCR‐RFLP techniques, we identified MTHFR gene polymorphisms (C677T & A1298C) associated with gestational diabetes.ResultsSignificant associations were found in gestational age and platelet count, indicating their relevance to GDM risk. The odds ratios for both MTHFR A1298C (p value: 0.034; OR = 0.7; 95% CI: 0.5021–0.9758) and MTHFR C677T (p value: 0.008; OR = 0.5453; 95% CI: 0.3292–0.9011) gene polymorphisms demonstrated an increased risk of GDM development.ConclusionThe study reveals that elevated platelet count and polymorphs increase the risk of developing GDM in pregnant women. Gestational age also plays a role. The study also finds a link between the MTHFR C677T SNP and the MTHFR A1298C gene polymorphism and the risk of gestational diabetes in Andhra Pradesh. This gives us new information about genetic and hematological factors.
  • Prevalence of resistant hypertension and its relation with plasma lipoprotein‑associated phospholipase A2

Background: A major modifiable risk factor for cardiovascular disease is hypertension. Resistant hypertension is observed to be associated with an increased likelihood of deleterious effects such as cardiovascular events, cerebrovascular accident, and renal dysfunction. The raised levels of inflammatory cytokines in the plasma like the C-reactive protein (CRP) have previously been observed to be significantly raised in hypertensive patients, but the overlying mechanisms corresponding to these processes remain unknown. Materials and Methods: In essence, this was an observational study with data collected from a population at a specific moment in the time to assess the relationships between the variables analyzed. A total of 200 individuals with arterial hypertension took part in the study. Fasting venous blood was obtained to evaluate plasma lipoprotein-related phospholipase A2 (Lp-PLA2) activity, and baseline data were collected. To diagnose RH, a 24-h ambulatory blood pressure management was done. Results: RH was observed among 100 patients and found to be predominant among males and older individuals, who were smokers and having hypertension for a longer period of time with higher Lp-PLA2 activity. More RH patients used calcium channel blockers and diuretics, while a fewer used angiotensin-converting enzyme inhibitors, angiotensin receptor blockers, and statins (P 0.05 for all comparisons). Conclusion: The plasma Lp-PLA2 activity beyond a certain threshold suggests a raised risk of RH, and the statins can help in reducing the RH incidence among persons with high Lp-PLA2 levels.
  • Association of TNFSF11 rs2200287 and TNFSF11 rs2148072 gene polymorphisms in preeclampsia

BACKGROUND Various cytokines released by white blood cells like lymphocytes are linked to the immune system during pregnancy. The polymorphism of the TNFSF11 (rs2200287 and rs2148072) gene is related to the preeclampsia of pregnancy. MATERIALS AND METHODS This study was a prospective study involving 304 pregnant women with preeclampsia (n = 152) and controls (non-preeclamptic pregnant women) (n = 152). To investigate the rs2200287 and rs2148072 SNP's of TNFSF11 gene polymorphism by using the PCR - RFLP techniques. RESULTS A significantly different genotype distribution of TNFSF11 (rs2200287 and rs2148072) polymorphisms were observed between the two groups, with the G allele of variant rs2200287 was highly significant in the preeclamptic group (P = 000; 0.5814; OR = 0.5814; 95% CI = 0.4211 - 0.8012). And the C allele of variant rs2148072 was also highly significant in the preeclamptic group (P = 000; OR = 0.5076; 95% CI - 0.362 - 0.71) in this study. CONCLUSION The outcomes of the present study indicate that there was an association in TNFSF 11 (rs2200287 and rs2148072) gene polymorphism with preeclampsia compared to non - preeclampsia women.
