Zyryanov, Mekala Nageswararao Chemical Biology and Drug Design, 2023 The privileged 1,2,3‐triazole scaffold is drawing researcher's attention due to its widespread applications in diverse fields such as drug discovery (e.g., carboxyamidotriazole), organic synthesis (click‐reaction template), polymeric materials (e.g., triazolamer), supramolecular receptors (e.g., triazolophane), fluorescent materials (e.g., metal–organic frameworks), and agricultural sectors (e.g., fungicides). Various 1,2,3‐triazole persuasion modules are also currently available in the market that have multiple assets such as active pharmaceuticals and agricultural purposes. Owed to the highly consistent and firmest synthesis approach, that is, click reaction of various azides and acetylene derivatives by copper (I)‐catalyzed 1,3‐dipolar cycloaddition (CuAAC), highly functionalized 1,2,3‐triazoles are prepared in scalar yields for drug discovery. Given the importance of 1,2,3‐triazole chemistry, the present review focuses specifically on the synthesis of structurally diverse 1,2,3‐triazoles linked to natural pharmacophores and their biological importance. Furthermore, the dual/multi‐pharmacophores assimilated 1,2,3‐triazoles have listed interesting biological activities that could be valuable as future drug leads. In addition, this comprehensive review can serve as a template for the development of new diverse scaffolds that will ensure for new therapeutic approaches for the existing myriad diseases and disorders.
Synthesis, Antimicrobial Assay and SARs of Pyrazole Included Heterocyclic Derivatives N. Mahaboob Basha, B.C. Venkatesh, Guda Mallikarjuna Reddy, Grigory V. Zyryanov, Munagapati Venkata Subbaiah, Jet-Chau Wen, Anjani R.K. Gollakota Polycyclic Aromatic Compounds, 2023 As pyrazoles are familiar for showing various biological properties, a series of novel isocoumarin tethered carbothioamide linked pyrazole derivatives were prepared successfully in the presence of environmental favor conditions and evaluated for their pathogenic resistance against four bacteria and two fungi, showing significant activity against different microbial except compound 5d. Among the active compounds, 5f displayed highest antimicrobial property. In fact, 5b and 5l motifs showing good inhibitory activity. Further, a detailed structure activity relationship also discussed, which revealed that the presence of electron withdrawing -NO2 group in 5f compound may delivered highest biological property, while the amino group attached in compound 5d delivered inactive antibacterial property. Moreover, computer aided studies of biologically effective compounds to determine the interactions and docking score with the crystal structure of Escherichia coli 24 kDa DNA gyrase subunit B in complex with clorobiocin. It was observed that the amino acid residues present in the active binding site of the protein1KZN can frequently interact with targets. Graphical Abstract

If those prepared compounds may possess therapeutically potentiality, then it called as efficient research. Herein, we report the 12 new oxadiazole linked carboxamide pyrazole substances have been synthesized and screened for their free radical scavenging activity by using hydrogen peroxide (H2O2), nitric oxide (N2O), and diphenyl picryl hydrazide (DPPH) methods. All the compounds displayed reliable biological properties. Among them, compound 8f hold high antioxidant activity may be due to the presence of methoxy group at para position of benzene ring. From the synthetic point of view, this is the novel environment favor method for the synthesis of pyrazole linked oxadiazoles, and the biological results of these compounds may stand as a referee for further development of biologically active pyrazole tethered oxadiazoles.

The compounds bisoxazolyl oxadiazole and bisoxazolyl thiadiazole having methyl substituent on the aromatic ring exhibited greater antioxidant activity than the standard ascorbic acid.
