In-vivo Wound Healing Activity of Acalypha paniculata , A. Elumalai, P. Shanmugapandiyan, , V. Vijay Kumar, , L. Kaviarasan, and Natural and Life Sciences Communications, 2025 The wound healing properties of the ethanol extract of dried aerial parts of Acalypha paniculata (APE) were investigated using excision and incision wound models in Wistar strain albino rats. In this experimental study, second-degree burn wounds were created in twenty prescreened rats, which were divided into four groups of five animals each. Group I served as the control and received normal saline, Group II was treated with povidone-iodine as the standard drug, and Groups III and IV were administered topically APE at doses of 250 and 500 µl/wound respectively. For the excision wound model, wound healing was assessed using parameters such as the percentage of wound contraction and epithelialization duration on days 2, 6, 10, and 14, along with histopathological evaluations. In both models, rats treated with APE demonstrated significantly enhanced wound healing activity (P <0.001) compared to the control group. This was evident from the accelerated wound contraction, shorter epithelialization period, increased skin tensile strength, and complete healing within 18 days for excision and seven days for incision method. Histopathological analysis further confirmed these findings, showing well-developed hair follicles, denser collagen fibers, reduced inflammatory cells, mature fibroblasts, and enlarged capillary-sized blood vessels, comparable to the outcomes observed in the standard treatment group. Our study demonstrates that the positive effects of APE on wound healing can be attributed to its ability to reduce wound size.
Stability Indicating Bioanalytical Validation of Elexacaftor, Ivacaftor and Tezacaftor using HPLC in Human Plasma S. Marakatham, P. Shanmugapandiyan Research Journal of Pharmacy and Technology, 2023 A simple, precised, accurate method was developed for the estimation of Elexacaftor, Ivacaftor and Tezacaftor in human plasma using the Lumacaftor as internal standard by RP-HPLC (Reverse phase-High performance Liquid Chromatographic) technique. Chromatographic conditions used are stationary phase Azilent (250 x 4.6mm, 5m), Mobile phase 0.01N Potassium di-hydrogen phosphate (pH: 3.5) : Acetonitrile in the ratio of 70:30(v/v) and flow rate was maintained at 1.0ml/min, detection wave length was 250nm, column temperature was set to 30oC and diluent was mobile phase Conditions were finalized as optimized method. Retention time of Ivacaftor, Elexacaftor and Tezacaftor were found to be 2.391min, 3.208min and 3.644min. %CV of the Elexacaftor, Ivacaftor and Tezacaftor was found to be 0.08%, 1.05% and 3.59%. %Recovery was obtained as 96.41%, 95.029% and 98.21%. The linearity concentration is in the range of 435-17400ng/mL of Elexacaftor, 60-2400ng/mL of Ivacaftor and 300-1200ng/mL of Tezacaftor (r2 = 0.999) .The lower limits of quantification were 435ng/mL of Elexacaftor, 600ng/mL of Ivacaftor and 300ng/mL of Tezacaftor which reach the level of both drugs possibly found in human plasma. Further, the reported method was validated as per the ICH guidelines and found to be well within the acceptable range. The proposed method is simple, rapid, accurate, precise, and appropriate for pharmacokinetic and therapeutic drug monitoring in the clinical laboratories.
Preliminary Phytochemical Analysis on The Leaves Extracts of Sida acuta Burm.f. and Sida rhombifolia Linn. Family Malvaceae Selvadurai S., Shanmugapandiyan P. Research Journal of Pharmacy and Technology, 2022 To investigate the phytochemical screening (group determination) of the plants leaves of Sida acuta Burm.f., and Sida rhombifolia Linn. family Malvaceae. In Ayurvedic system of medicine Sida acuta Burm.f., used as digestive, diuretic, fever, burning of the body and urinary discharge. Sida rhombifolia Linn. used as good in urinary complaints, discharge and strangury, useful in fever, heart diseases, burning sensations, piles, all kinds of inflammations. Present study involved Leaves of Sida acuta Burm.f., and Sida rhombifolia Linn. were studied by preliminary phytochemical screening of leaf powders using different solvent viz; petroleum ether, chloroform, acetone, ethanol and aqueous. Leaf extracts were subjected to preliminary phytochemical analysis of using standard procedures. The phytoconstituents present in it viz. alkaloids, carbhohydrates, glycosides, phytosterols, phenolic compounds, tannins, flavonoids, lignins and saponins are present in leaf extracts of Sida acuta Burm.f., and Sida rhombifolia Linn. The active principles of Sida acuta Burm.f., and Sida rhombifolia Linn. namely Manghaslin, Quercetin 3-(2Gxylosylrutinoside), Myricetin 7-Rhamnoside, Isorhamnetin 3-O-[b-D-glucopyranosyl-(1->2)-a Lrhamnopyranoside], Quercetin 3-(2Gxylosylrutinoside), Peltatoside, Rutin, Quercimeritrin are present. The diversity of phytochemicals study can serve as a valuable source of information and provide suitable standards for identification of this plant material in future investigations.
Bioanalytical method development and validation of doravirine, lamavudine and tenofovir disoproxil fumarate using hplc in human plasma Marakatham S., Shanmugapandiyan P. Research Journal of Pharmacy and Technology, 2021 A novel, simple and sensitive bioanalytical method was developed for estimation of Doravirine, Lamavudine and tenofovir disoproxil fumarate in human plasma with daclatasvir as internal standard. The method was developed using alliance HPLC using Phenomenex C18 (150mm x 4.6mm, 5m) column with mobile phase of 0.01N Potassium dihydrogen phosphate pH (3.5): Acetonitrile (60:40) at flow rate of 1.0ml/min. Detection wavelength was found to be 277nm. The linearity range for doravirine, lamuvidine and Tenfovir was 50-2000ng/ml, 125-5000ng/ml and 20-800ng/ml. Correlation coefficient was 0.999. The method was validated and stability study was carried out as per FDA guidelines.
Combined effect of levothyroxine and oral hypoglycemica agents on blood glucose level in hypothyroidism patients-a prospective observational study Anandkumar S., Shanmugapandiyan P. International Journal of Research in Pharmaceutical Sciences, 2020 This study was aimed to evaluate the effect of levothyroxine combination therapy on blood glucose level in hypothyroidism patients. The Study was carried out in a private hospital at Tiruchengode as prospective observationally. Patients who were under the management of thyroid disorder and diabetes mellitus for 1 year period was screened and selected for the study. Effect of levothyroxine on blood glucose level and using a Mini mental state examination questionnaire their cognitive skills were also assessed. Independent sample t' test was used to compare the blood glucose level and complication of the management. There were 42 patients were included in the study and the patients who were on Levothyroxine and Glimepride with Metformin labeled as Group 1 and Glimepride with Metformin alone was labeled as Group 2. Glimepride with metformin therapy shows greater percentage of blood glucose level reduction and significant p<0.05 cognitive impairment was found in the combination therapy. Diabetes mellitus management with levothyroxine and glimepride with metformin has an association on the managing blood glucose level. Efficient management of blood glucose level was obtained with glimepride with metformin alone. Cognition impairment was high in the combination therapy of levothyroxine and glimepride with metformin.
Clinical investigation on mini mental score assessment on various stages of hypothyroidism patients S. Anandkumar, P. Shanmugapandiyan Research Journal of Pharmacy and Technology, 2019 To assess the cognitive impairment in various conditions of hypothyroidism patients. The study design was a cross sectional observational case control study. The study site was carried out in the department of general medicine at Vivekanandha medical care Hospital, Tiruchengode. Patients of subclinical hypothyroidism without neurological disorder, Patients of overt hypothyroidism without neurological disorder, Normal Euthyroid persons were included in the study, Pediatrics Patients, Comatose patients, Chronic Illness patients, Patients with neurological disorder, pregnant women were excluded from the study. The average MMSE score of hypothyroid patients was 22.56±3.36, average MMSE score of control group is 24.94±2.48. When compared the both group shows (p <0.0001) extremely significant. Hypothyroidism patients have low MMSE score than euthyroid participants which reveals hypothyroidism is significantly associated with cognitive impairment. Present study also reveals patients who have chronic thyroid damage leads to higher risk of cognitive impairment.
