    Elsevier BV
Abstract Our study aimed to prepare nanoemulsions coated with alginate/Aloe vera gel (A/AV) for oral insulin delivery. Uncoated nanoemulsions were prepared by homogenization of water in oil in water (w/o/w) multiple emulsions that were composed of castor oil (emulsifier). Coating of the nanoemulsions was achieved based on ionic gelation method, with sequential addition of calcium chloride and Aloe vera gel (AV) to the bulk nanoemulsion dispersion that contained alginate. The particle size of the coated nanoemulsions was about 400 nm and the insulin entrapment ratio was 47.3%. In vitro leakage study indicated well-preserved integrity of the nanoemulsions in simulated gastric juices. In conclusion, the nanoemulsion coated with alginate/Aloe vera gel (A/AV) was a potential delivery system for oral delivery of polypeptides and proteins. Feasibility of this nanoemulsion for oral delivery of insulin (model peptide) is investigated in Caco-2 cell line using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide viability assay. The cell culture results for translocation of insulin across the cell monolayer are very promising (20%–25% increase), and animal studies are actively under progress and will be published separately in near future.

  • Polysaccharides as excipient in drug delivery system
    S. Khaleel Basha, M. Syed Muzammil, R. Dhandayuthabani, and V. Sugantha Kumari

    Elsevier BV
Abstract This review examines the large polysaccharide particles play in functional drug delivery. The significance of these frameworks is because of the wide assortment of polysaccharides and their typical source which implies that they can achieve biocompatible and biodegradable systems with a scope of biological and chemical usefulness benefits critical to drug tracking / delivery. This value incorporates giving and presenting specialists working through shirk of the reticuloendothelial framework, regulating bio-macromolecules, and growing the bioavailability of incorporated small molecule drugs. Transport of the helpful is likewise key basic for the utilization of polysaccharide particles, moves prescriptions from the site of organization through mucosal official and transport, and utilizing the science, size and receptor meddles sedate focusing to explicit tissues. This review also elaborates on techniques for integrating and developing frameworks for the use of high-affinity sedate transport polysaccharide molecules and extending the usefulness of normal polysaccharides.
