  • From ecology to evolution: plasmid- and colicin-mediated persistence of antibiotic-resistant Escherichia coli in gulls

    Antimicrobial resistance (AMR) in wildlife is an emerging concern within the One Health concept. Gulls, due to their synanthropic behavior and long-distance migration, are recognized as vectors and secondary reservoirs of resistant bacteria. These birds can facilitate the environmental spread of resistant strains across ecosystem boundaries. Understanding their role in shaping microbial communities is essential for assessing the broader ecological impact. This study investigates the persistence and competitive dynamics of cephalosporin-resistant Escherichia coli in Caspian gulls ( Larus cachinnans ) captured at their breeding colony at a water reservoir and subsequently monitored in captivity for three months, representing the longest in vivo experiment of its kind conducted on wild birds. We observed sustained colonization and long-term shedding of resistant E. coli throughout the entire study, marking the longest documented carriage of resistant bacteria in wild birds to date. Notably, rapid dissemination of various E. coli sequence types (STs) with CTX-M-1 was observed, with ST11138 rapidly outcompeting other strains, including the initially dominant ST11893. Genomic analyses revealed that ST11138 harboured F24:A-:B1 and IncI1/ST3/CTX-M-1 plasmids encoding colicins and corresponding immunity genes, likely conferring a competitive advantage. Our findings underscore the role of bacteriocin-mediated interactions in shaping microbial communities and highlight the importance of plasmid-encoded traits in the persistence of resistant strains in wildlife. Importantly, our findings underscore the ecological novelty of longitudinal in vivo tracking of AMR persistence in natural hosts and highlight the need to consider ecological and microbiome-level interactions when assessing the environmental dimension of AMR under the One Health concept. However, the role of wildlife in resistance dynamics remains underexplored, especially in long-term, natural settings. Our study is unique in its scope and duration, representing the longest in vivo experiment of its kind conducted on wild birds. By capturing these processes in live hosts under naturalistic conditions and across an extended period, our study provides rare and ecologically grounded insights into how AMR is maintained outside clinical or laboratory settings. Our findings show sustained colonisation and long-term shedding of resistant E. coli , with strain ST11138 outcompeting others. Genomic analyses reveal plasmid-encoded traits, highlighting the ecological and evolutionary mechanisms underlying resistance maintenance in wildlife.
  • Generation of Bioactive Stem Cell-Derived Secretome in 3D Bioreactor System: Towards Cell-Free Therapy in Veterinary Medicine

    Canine adipose-derived mesenchymal stem cells (cASC) are promising for regenerative veterinary medicine due to their immunomodulatory and reparative capacities. Three-dimensional (3D) culturing provides a more physiologically relevant environment than conventional two-dimensional (2D) monolayers, enhancing paracrine activity and therapeutic potential of mesenchymal stem cells (MSC). This study investigates the production and biological characterization of cASC secretome generated under hypoxic conditions with platelet lysate (PLT) supplementation, either in a 2D culture or in a stirred-tank 3D culture. Secretomes obtained from 3D cultures were compared with those from 2D cultures prepared under identical hypoxic and PLT-supplemented conditions. Quantitative analyses revealed enhanced secretion of key factors, including monocyte chemoattractant protein-1 (MCP-1) and vascular endothelial growth factor (VEGF), in 3D-derived secretomes. Functional in vitro assays demonstrated superior anti-inflammatory, pro-migratory, and antifibrotic effects of the 3D secretome, evidenced by nuclear factor kappa B (NF-κB) inhibition, increased fibroblast migration, and modulation of extracellular matrix gene expression. Additionally, the bioreactor system enabled consistent secretome production with reproducible biological activity. These findings indicate that 3D bioreactor cultivation under hypoxia with PLT supplementation can generate a biologically active secretome from canine adipose-derived stem cells, providing a promising basis for further exploration in veterinary regenerative applications.
  • Growth stage and interspecies interactions shape the cell biology and cell cycle characteristics of human gut bacteria Bacteroides thetaiotaomicron and Roseburia intestinalis

    The human gut is a densely populated environment where microbial cells face frequent nutrient fluctuations. While previous studies have identified changes in gene expression, protein concentrations, and metabolite levels, less is known about how gut bacteria respond to nutrient fluctuations at the cellular level. Here, we compared cell biological properties of B. thetaiotaomicron and R. intestinalis, two prevalent human gut microbiome members. Our comparison revealed distinct cellular properties of both bacteria during exponential growth in vitro, including a unique zonal peptidoglycan synthesis pattern in R. intestinalis. Upon nutrient depletion in stationary phase, we observed different cell morphological alterations, with B. thetaiotaomicron cells becoming wider and R. intestinalis cells shorter, accompanied by distinct intracellular changes. These alterations extended to other B. thetaiotaomicron strains, but not Roseburia species. By re-analyzing an existing RNA-seq dataset, we could link the cell biological response of both species to different alterations in gene expression in stationary phase. Finally, coculture experiments revealed a significant impact of interspecies interactions on cell morphological and transcriptomic properties of both bacteria. Together, our work provides insight into the unexplored cell biology of representative human gut microbiome members and how this is shaped by nutrient fluctuations and the presence of other species.
  • Novel Insights into Systemic Hyaluronic Acid Therapy in Dogs with Osteoarthritis from an Exploratory Postmarketing Study: Clinical Improvements Linked to Biomarker Changes

    This prospective, single-arm, exploratory postmarketing study preliminarily evaluated the clinical response and plasma biomarker changes in 18 client-owned dogs with naturally occurring osteoarthritis (OA) treated with sodium hyaluronate (Bonharen). Patients received intravenous injections of Bonharen Intravenous at a dose of 0.15 mL/kg (1.3–1.6 mg/kg hyaluronic acid once a week for consecutive five weeks). Clinical parameters (lameness, joint pain, mobility, swelling) were assessed weekly and two weeks after the final dose was given via standardized scoring. The plasma concentrations of selected inflammatory, cartilage-related, and oxidative stress biomarkers were measured before treatment and two weeks after the final dose. Clinical improvement in lameness and/or joint pain on palpation was observed in nearly half of the patients. No clinical deterioration was recorded at any time point. Physical activity increased in all patients with reduced baseline activity. Significant decreases in the plasma levels of prostaglandin E2, Δ17-6-keto prostaglandin F1α, malondialdehyde, and hyaluronan were detected, indicating reduced systemic inflammation and oxidative stress. In addition, an increase in plasma hydroxybutyrate and decrease in the collagen-breakdown marker prolyl-hydroxyproline were observed. No adverse effects were reported. These findings suggest that intravenous hyaluronic acid (Bonharen) may represent a safe and promising component to multimodal OA management in dogs and demonstrate the feasibility of integrating plasma biomarkers in canine OA studies.
  • Intravenous Administration of Hyaluronan in Veterinary Musculoskeletal Disorders: Therapeutic Applications and Mechanisms

    Hyaluronan (HA), a natural polysaccharide, is a significant component in the extracellular matrix of connective tissues. Recently, HA has gained substantial attention in veterinary medicine due to its remarkable effects in promoting wound healing and supporting joint health. The route of administration plays a key role in the therapeutic potential of HA, as it affects the pharmacokinetics and bioavailability of HA within the body and diseased joints. Moreover, administration routes differ considerably regarding technical complexity and patient discomfort. This review summarizes recent studies exploring therapeutic applications of intravenous HA across various medical conditions, animal species, and clinical settings, emphasizing specific therapeutic effects, underlying mechanisms, clinical efficacy, and overall therapeutic potential, highlighting systemic effects beneficial for multijoint and systemic connective tissue conditions.
  • Alleviating UV-induced photoaging with oral hyaluronic acid: insights from a hairless mouse model

    Hyaluronic acid (HA) is known for its beneficial effects on skin hydration and elasticity. However, the mechanisms by which HA dietary intake attenuates pathological processes in photoaged skin are still unclear. In this study, oral administration of HA, alone or in combination with hydrolysed collagen, was investigated to alleviate UV-induced inflammation, extracellular matrix (ECM) degradation, and metabolic alterations in the skin of SKH-1 hairless mice repeatedly exposed to UVA and UVB radiation. Both supplements reduced UV-induced degradation of HA and collagen and decreased the inflammatory response in the skin. Interestingly, HA supplementation influenced cutaneous and plasma metabolism, elevating lipid catabolism and metabolic markers such as hydroxybutyrate and a-ketoglutarate. The addition of collagen did not provide any additional significant benefits over HA despite being administered at a 12-fold higher dose. These findings highlight superior role of HA as a nutritional supplement for skin health and provide insights into its potential mechanisms in addressing photoaged skin by modulating both skin and systemic metabolism.
  • Complex interplay between gene deletions and the environment uncovers cellular roles for genes of unknown function in Escherichia coli

    Phenotypic outcomes can be heavily affected by environmental factors. In this study, we exploited the previously observed nutrient dependency of cell biological phenotypic features captured by a cross-condition image-based profiling assay of Escherichia coli deletion strains to examine this in more detail. We identified several general principles, including the existence of a spectrum of deviating phenotypes across nutrient conditions (i.e., from nutrient- or feature-specific to pleiotropic phenotypic deviations), limited conservation of phenotypic deviations across nutrient conditions (i.e., limited phenotypic robustness), and a subset of nutrient-independent phenotypic deviations (indicative of consistent genetic determinants of specific phenotypic features). In a subsequent step, we used this cross-condition data set to identify five genes of unknown function ( yibN , yaaY , yfaQ , ybiJ , and yijD ), of which the deletion displayed either nutrient-independent phenotypic deviations or phenotypic similarities to genes of known function. These genes showed different levels of phylogenetic conservation, ranging from conserved across the tree of life ( yibN ) to only present in some genera of the Enterobacterales ( yaaY ). Analysis of the structural properties of the proteins encoded by these y-genes, identification of structural similarities to other proteins, and the examination of their subcellular localization yielded new insights into their contribution to E. coli cell morphogenesis, cell cycle progression, and cell growth. Together, our approach showcases how bacterial image-based profiling assays and data sets can serve as a gateway to reveal the function of uncharacterized proteins. IMPORTANCE Despite unprecedented access to genomic information, predicting phenotypes based on genotypes remains notoriously difficult. One major confounding factor is the environment and its ability to modulate phenotypic outcomes. Another fact is that a large fraction of protein-coding genes in bacterial genomes remain uncharacterized and have no known function. In this work, we use a large-scale cross-condition image-based profiling dataset to characterize nutrient-dependent phenotypic variability of Escherichia coli deletion strains and exploit it to provide insight into the cellular role of genes of unknown function. Through our analysis, we identified five genes of unknown function that we subsequently further characterized by examining their phylogenetic conservation, predicted structural properties and similarities, and their intracellular localization. Combined, this approach highlights the potential of cross-condition image-based profiling, which extracts many cell biological phenotypic readouts across multiple conditions, to better understand nutrient-dependent phenotypic variability and uncover protein function.

  • Optimizing Hyaluronan-Based Lubricants for Treating Thoracolumbar Fascia Pathologies: Insights from Tribological and Pharmacokinetic Studies

    In a world where the incidence of non-specific lower back pain (LBP) is steadily increasing, researchers are still searching for effective solutions for patients. Hyaluronic acid (HA) viscosupplementation is commonly used to restore lubrication in osteoarthritis (OA) and other medical applications, but its rapid metabolism limits efficacy. This study evaluates whether an HA derivative can replace native HA for the treatment of non-specific LBP while maintaining or enhancing its frictional properties and improving in vivo stability. Six HA-based lubricants, both native and derivatized, were tested in a tribological rabbit fascia model and a new synthetic model. Reduced HA derivative showed better tribological properties and longer in vivo residence time compared to native HA, as demonstrated in pharmacokinetic studies in rabbits. The 316 kDa HA and reduced HA exhibited the most stable tribological properties, which were influenced by their molecular weight and concentration. These findings suggest that both native and reduced HA are promising viscosupplements for intrafascial injection in the treatment of LBP, with reduced HA potentially enhancing effectiveness through a prolonged effect.
  • In situ delivery of synthetic preimplantation factor using aldehyde-modified hyaluronic acid hydrogel with immobilized complexes of chondroitin sulfate derivatives

    Synthetic preimplantation factor (SPIF) is a promising therapeutic agent for chronic inflammatory diseases like Multiple Sclerosis (MS), but frequent systemic dosing limits patient adherence and therapy efficacy. This study presents an injectable drug delivery system (DDS) using 2 % (w/v) aldehyde-modified hyaluronic acid (HAOX) and chondroitin sulfate (CSOX) to deliver 100 µg of Fluorescein isothiocyanate-modified SPIF (FITC-SPIF). The DDS utilizes electrostatic interactions between the negatively charged sulfate groups of CSOX and the positively charged amino acids of FITC-SPIF for effective entrapment. Key properties were analyzed, including moderate gelation time (193 s), swelling profile (18 %), injectability (27 G needle) and an established relevant release profile (20 μg/daily ± 3) via anomalous diffusion. Increasing CSOX concentration reduced initial burst release by 38 % (0.5 % CSOX) to 78 % (1 % CSOX), extending release time (T50 %) from 50 h (0.5 % CSOX) to 88 h (1 % CSOX). Additionally, the release of FITC-SPIF enhanced TGF-β secretion in THP-1 macrophages, indicating preserved biological activity. These findings highlight the tunable release and mechanical properties achieved by adjusting the HAOX:CSOX ratio, strategically aligning the DDS for targeted MS symptom management. This system potentially simplifies SPIF delivery and enhances therapeutic efficacy.
