Computational identification of multi-target natural compounds from Sesbania grandiflora as potential therapeutic agents against Klebsiella pneumoniae Harshit Sajal, Aswin Mohan, Vishal Ravi, K. Nadh Scientific Reports, 2026 Klebsiella pneumoniae (K. pneumoniae) is a Gram-negative bacterium that causes severe community- and hospital-acquired infections. Its rising multidrug resistance complicates therapy, highlighting the need for novel drugs with broad-spectrum, multi-target potential. Leveraging the traditional use and therapeutic evidence of Sesbania grandiflora, this study performed structure-based computational screening of its phytochemicals against K. pneumoniae targets. Initially, 93 proteins with high annotation scores and resolved X-ray structures were identified. Six key therapeutic targets, including LpxH, fabG, KPC-2, GlmU, chbG, and ompA, were prioritized for their pathogenic role. Molecular docking revealed that 59 of 73 compounds interacted with all six targets with high affinity, while the remaining 14 compounds interacted with five targets. Network pharmacology indicated KPC-2, fabG, and ompA had the highest connectivity (73 compounds), followed by chbG and LpxH (72), and GlmU (61). ‘3′,6-di-O-feruloylsucrose’ had the strongest affinity for ompA, LpxH, and GlmU, while ‘Acarbose hydrate’ ranked top for chbG, fabG, and KPC-2. Out of 47 drug-like compounds, 9 passed ADMET filters. Sonchuionoside A was selected for molecular dynamics simulations, demonstrating stable binding to all targets. This suggests S. grandiflora phytoconstituents as multi-target regulators against K. pneumoniae and highlights Sonchuionoside A as a promising lead for further validation.

Rajeev, Rajesh Raju Discover Oncology, 2026 ARID1A, a subunit of the SWI/SNF chromatin-remodeling complex, plays a critical role in maintaining genomic stability and regulating estrogen receptor (ER) signaling, yet its phosphorylation dynamics in cancer remain underexplored. This study employed phosphoproteomic analysis to investigate ARID1A phosphorylation in breast cancer, identifying predominant phosphosites—S363, S1184, and S696—regulated by kinases such as MAPK14, CDK16, and MAPK9. Functional enrichment revealed ARID1A interactions with SWI/SNF components (e.g., PBRM1, SMARCC2, BRD9) and DNA damage response (DDR) proteins (e.g., TP53BP1, TOP2A, CHEK2, NBN), underscoring its dual role in chromatin remodeling and double-strand break repair. Notably, phosphorylation at S363 and S1184 was significantly upregulated in breast cancer, suggesting tumour-specific hyperphosphorylation that may disrupt ARID1A tumour-suppressive function and contribute to endocrine resistance. Dysregulation of these phosphorylation events correlated with enhanced MAPK signaling, cancer progression, and poor prognosis. These findings position ARID1A as a molecular hub linking chromatin dynamics to genome integrity, with implications for therapeutic resistance. Targeting ARID1A phosphorylation pathways, potentially via MAPK inhibitors or BRD4/BRD9 antagonists, could restore its suppressive activity and improve treatment outcomes in breast cancer. This study enhances our understanding of ARID1A regulatory mechanisms, highlighting its phosphorylation as a key driver of breast cancer biology. Future research should validate these kinase-substrate interactions and explore their transcriptional and chromatin-level impacts to develop precision therapies for ARID1A-dysregulated cancers.
Evaluation of phytochemicals from Tamarindus indica as a potential catechol-O-methyltransferase (COMT) inhibitor: an in-silico approach for Parkinson’s disease Aparna G. Dopamine is biosynthesized from L-tyrosine and is primarily metabolized by catechol-O-methyltransferase (COMT), which regulates its concentration and ensures proper synaptic signalling within the basal ganglia. Dopamine deficiency, the hallmark of Parkinson’s disease, is a progressive neurodegenerative disorder caused by the loss of dopaminergic neurons in the substantia nigra pars compacta. To alleviate dopamine deficiency, current therapeutic strategies include administration of the dopamine precursor levodopa in combination with COMT inhibitors, which enhance levodopa bioavailability and prolong its therapeutic effect. However, the clinical utility of synthetic COMT inhibitors is often limited by hepatotoxicity and other adverse effects, underscoring the need for safer alternatives. In this context, plant-derived bioactive compounds are being increasingly investigated as potential inhibitors of COMT activity. Our study evaluated the affinity of 170 phytochemicals from Tamarindus indica against COMT utilizing computational strategies. Gallacetophenone, an aromatic ketone, was identified as a promising candidate that met the requirements for drug-likeness. Molecular dynamics (MD) simulations, supported by post-MDS analyses, principal component analysis, and free energy landscape analyses, demonstrated that gallacetophenone binding enhances the conformational stability of 3BWY compared to the control. These findings suggest that gallacetophenone may serve as a potential therapeutic phytochemical; however, its efficacy remains computationally predicted and requires validation through targeted in vitro and in vivo studies.
Integrative phosphoproteomic analysis reveals co-regulatory phosphorylation networks of rhotekin in cancer progression Akhina Palollathil, Althaf Mahin, Athira Perunelly Gopalakrishnan, Alimath Sambreena, Samseera Ummar, Prathik Basthikoppa Shivamurthy, R. Ganesan, Manavalan Vijayakumar, Rajesh Raju Discover Oncology, 2026 Rhotekin (RTKN), a Rho GTPase effector, promotes the development of malignancies, including breast, gastric, and colon cancers, by enhancing cell proliferation and migration while inhibiting apoptosis. Despite its oncogenic role, the phosphoregulatory network of RTKN remains largely unexplored, with no experimental evidence on its upstream kinases and functional phosphosites. To characterize RTKN-associated phosphorylation dynamics, PubMed-indexed studies were systematically retrieved using predefined MeSH terms to compile large-scale cellular phosphoproteomics datasets. Analysis of 618 quantitative profiling and 179 differential abundance datasets identified 27 Class-I phosphosites in RTKN. Among these, five sites-Ser106, Ser220, Ser520, Ser529, and Ser543 were consistently observed across multiple datasets, suggesting them as predominant sites with potential functional significance. Structural mapping of predominant sites onto the AlphaFold2-predicted model indicated that these sites are located in accessible regions, highlighting their potential susceptibility to kinase-mediated regulation. As these sites represent understudied phosphosites, a robust strategy was employed to identify their functional role by assessing co-regulated phosphosites on other proteins (PsOPs). ACIN1_Ser243, CTNNA1_Ser641, and SHROOM2_Ser1036 were among the top positively co-regulated PsOPs, whereas MICALL1_Ser644, PRP4K_Ser366, and MYO18A_Ser1970 were negatively co-regulated. PsOPs were mainly involved in apoptosis, cell growth, motility, and cytoskeletal reorganization, suggesting potential functional convergence with RTKN. Additionally, phosphorylation at RTKN Ser520 and Ser543 co-occurred across multiple datasets. Moreover, TRPM7 and PAK4 were identified as predicted upstream kinases phosphorylating RTKN at Ser220 and Ser520. Pathway analysis showed involvement of co-regulated proteins in cancer-associated signaling pathways. These findings provide a foundation for future research to elucidate the phosphosite-specific role of RTKN in cancer.
