    Antimicrobial resistance remains one of the major challenges to global public health, compromising the effectiveness of treatments and contributing to increased morbidity and mortality associated with bacterial infections. Among the mechanisms involved, efflux pumps—such as NorA, expressed in resistant strains of Staphylococcus aureus —are particularly noteworthy. These transport proteins actively expel antibiotics from the cell, reducing their intracellular concentration. In this context, natural compounds have been explored as potential resistance inhibitors, with a focus on the triterpene lupeol, known for its pharmacological properties. This study evaluates the activity of lupeol against the NorA efflux pump using in vitro assays and in silico modeling. The minimum inhibitory concentration (MIC) is determined by broth microdilution, and pump inhibition is assessed via ethidium bromide‐induced fluorescence. SYTOX Green assays indicated that lupeol does not compromise bacterial membrane integrity. Although lupeol presented a MIC ≥ 1024 μg mL −1 , it demonstrates significant inhibition of NorA activity. Molecular docking reveals a binding energy of –7.112 kcal mol −1 and interactions with key residues of the protein, outperforming the CCCP control. These findings suggest that lupeol acts as a modulator of bacterial resistance, with potential application as a therapeutic adjuvant in the treatment of infections caused by multidrug‐resistant S. aureus.

    Antimicrobial resistance is an increasing threat to public health, with alarming estimates of mortality rates. In this context, the clinical properties of Varronia curassavica Jacq. are highlighted due to its biological and pharmacological activities. This study aims to analyze the phytochemical composition of the essential oil from V. curassavica (EOVC) and its antibiofilm activity against Gram‐positive and Gram‐negative strains. The essential oil was extracted from the leaves using hydrodistillation and analyzed by gas chromatography–mass spectrometry (GC–MS), phytochemical analysis of EOVC identified 97.36% of the total composition, predominantly composed of monoterpenes (α‐pinene at 44.46%) and sesquiterpenes (β‐caryophyllene at 21.82%). Antibiofilm tests were performed on five bacterial strains (both Gram‐positive and Gram‐negative) to evaluate the ability to inhibit and eradicate biofilms, with statistical analysis of the results. Antibiofilm formation tests demonstrated that EOVC exhibited efficacy against Staphylococcus aureus and Staphylococcus epidermidis , with minimum inhibitory concentrations (MIC) of 1.024 and 10.240 µg/mL, respectively, values comparable to those observed for the reference antibiotic. In addition, at high concentrations (10 × MIC), EOVC inhibited biofilm development by Streptococcus mutans and Enterococcus faecalis , achieving performance similar to that of gentamicin. In biofilm eradication assays, EOVC showed effective activity against E. faecalis , S. epidermidis , and S. aureus at concentrations of 10.240 µg/mL, equivalent to gentamicin. However, resistance was found in strains of Pseudomonas aeruginosa and S. mutans , with P. aeruginosa showing the highest level of resistance to EOVC.

    Spondias purpurea L. is widely used in folk medicine to treat various pathological conditions. One of its promising activities is antimicrobial. The present work aimed to characterize the composition and evaluate the antifungal and antibacterial effects of aqueous (AESP) and ethanolic (EESP) extracts of leaves. The technique of ultra‐performance liquid chromatography coupled to a quadruple time‐of‐flight system (UPLC‐QTOF) was used for phytochemical qualification. For the antifungal tests, two strains of Candida albicans were used, and for the antibacterial tests, strains of Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus were used. Quercetin, Kaempferol, Quinic Acid, gallo (epi)catechin dimer, dehydroascorbic acid, and quercetin derivative were detected in AESP, and quinic acid, iriflofenone 3‐C‐β D glycopyranoside, chlorogenic acid, and stearidonic acid in EESP. It was found that AESP potentiated the action of amikacin against bacteria, and an antifungal effect of both extracts was observed at a concentration of 8,192 µg/mL for the CA URM 4127 strain. The two evaluated extracts significantly potentiated the effect of fluconazole against CA INCQS 40006. The research showed that the extracts tested alone do not present significant antibacterial or antifungal activities at low concentrations against the tested strains. However, they demonstrated potentiating effect of already standardized drugs.

    ABSTRACT Bacterial biofilms are structured communities of microorganisms embedded in an extracellular matrix, which confer high resistance to antibiotics and complicate the treatment of infections. In this context, the search for new strategies to control bacterial resistance becomes urgent. This study aimed to evaluate the susceptibility of planktonic cells and bacterial biofilms to the hydroethanolic extracts of the bark (HEBSJ) and leaves (HELSJ) of Sarcomphalus joazeiro , as well as to perform their chemical characterization. The extracts were obtained by cold maceration and analyzed using UPLC-ESI-QToF-MS E . Antibacterial activity and biofilm formation/inhibition assays were conducted in 96-well microdilution plates, with quantification performed using the crystal violet method and absorbance readings taken at 570 nm. All tests were performed in triplicate. Chemical analysis revealed the presence of flavonoids, organic acids, and saponins. Regarding biofilm-forming capacity, Staphylococcus aureus and Staphylococcus epidermidis were classified as strong biofilm producers; Enterococcus faecalis , Streptococcus mutans , and Pseudomonas aeruginosa showed moderate biofilm formation, while Escherichia coli was classified as a non-biofilm-producing strain. HELSJ exhibited inhibitory activity only against P. aeruginosa , while it appeared to promote biofilm formation in the remaining strains. In contrast, HEBSJ demonstrated inhibitory activity in all analyzed strains at least at one tested concentration, including complete inhibition of E. faecalis biofilm, outperforming chlorhexidine. In conclusion, the bark extract (HEBSJ) showed superior and consistent antibiofilm activity across all tested strains, with significant results compared to the leaf extract (HELSJ), which exhibited limited efficacy. These findings reinforce the need for further studies to elucidate the mechanisms of action involved and support the development of novel phytotherapeutic agents for the treatment of biofilm-associated infections.

    Biofilms, formed by microbial communities that increase resistance to antibiotics, are responsible for chronic infections, making their combat a therapeutic priority. Taking this into account, the fruit Caryocar coriaceum stands out for its potential in the treatment of infectious diseases. The different parts of this plant can be used, and the fixed oil extracted from its fruit, rich in fatty acids, is indicated as responsible for its biological activities. Thus, the objective of this study was to evaluate the chemical composition of the fixed oil extracted from the fruits of C. coriaceum (FOCC), in addition to analyzing its action in the inhibition and pre-formed biofilm disruption of bacteria. The fixed oil was extracted from the internal mesocarp through exhaustive extraction with n-hexane, resulting in a yield of 38.29%. For antibiofilm evaluation, multidrug-resistant bacterial strains were exposed to the oil, and the antibiofilm activity was verified through biofilm formation and pre-formed biofilm disruption assays. The chemical analysis of the fixed oil of C. coriaceum (FOCC) identified eight fatty acids, representing 98.2% of the total composition, with a predominance of oleic acid (60.1%) and palmitic acid (33.5%). FOCC demonstrated approximately 70% inhibition of Streptococcus mutans biofilm formation at a concentration of 10 mg/mL and approximately 60% inhibition against Staphylococcus aureus and Pseudomonas aeruginosa. In pre-formed biofilm disruption, FOCC showed low efficacy against S. mutans and P. aeruginosa but showed greater activity against Enterococcus faecalis and S. aureus. These results indicate that FOCC has the potential to prevent biofilms, but its pre-formed biofilm disruption capacity is still limited.
