- Structural elucidation of a crystalline mannan from açaí (Euterpe oleracea Mart.) seeds

Açaí is an Amazonian “superfruit” whose pulp production generates large amounts of seed waste. As nearly half of the dry mass of this residue is composed of mannan, this study investigated the structure and distribution of mannan in açaí seeds to support its bioconversion into value-added products such as mannose and mannan-oligosaccharides. To this end, the seeds were fractionated to obtain soluble and water-insoluble fractions, which were analyzed to assess their mannan content. In soluble fractions, glycomic profiling revealed the presence of an alkali-soluble mannan together with low amounts of pectin, (hetero)xylan, xyloglucan, and glucan. The water-insoluble fraction was further analyzed by solution-state NMR, where 1 H/ 13 C HSQC and HMBC spectra showed signals consistent with a backbone of 1,4-linked β-D-Man p , with chemical shifts similar to those reported for mannan from ivory nut, a material known to contain linear mannan. Gas chromatography analysis indicated a predominantly linear mannan with mannose-to-galactose ratio of 50:1, suggesting only minor galactose substitutions. Furthermore, due to the limited solubility of this fraction, 13 C CP/MAS solid-state NMR was employed to enable a comprehensive structural assessment. Solid-state NMR revealed the presence of a mannan I polymorph, with its crystalline nature further confirmed by X-ray diffraction. The identification of a highly crystalline linear mannan in açaí seeds provides structural insight relevant to the development of strategies aimed at reducing mannan crystallinity and designing specialized enzymatic cocktails for its efficient hydrolysis, thereby supporting the sustainable valorization of this abundant agro-industrial byproduct. • Glycome profiling identified epitopes of xyloglucan, xylan and an alkali-soluble mannan in seeds. • There is an insoluble linear β-(1 → 4)-D-mannan structure in açaí seeds endosperm. • Solid-state NMR and X-ray diffraction confirmed a crystalline mannan I polymorph. • Crystallinity is a structural feature that may contribute to mannan recalcitrance. - GlycoGenius: a streamlined high-throughput glycan composition identification tool

Mass spectrometry is recognized as the gold standard for glycan analysis, yet the complexity of the generated data hampers progress in glycobiology, as existing tools lack full automation, requiring extensive manual effort. We introduce GlycoGenius, an open-source program offering an automated workflow for glycomics data analysis, featuring an intuitive graphical interface. With algorithms tailored to reduce manual workload, it allows for data visualization and automatically constructs search spaces, identifies, scores, and quantifies glycans, filters results, and annotates fragment spectra of N- and O-glycans, glycosaminoglycans and more. It seamlessly guides researchers of all expertise levels from raw data to publication-ready figures. Our findings demonstrate that GlycoGenius achieves results comparable to manual analysis or competing tools, identifying more glycans, including novel ones, while significantly reducing processing time. This groundbreaking tool represents a significant advancement in the study of glycoconjugates, empowering researchers to focus on insights rather than data processing. This study shows how β- O-linked- N-acetylglucosamine ( O-GlcNAcylation) modulates tyrosine hydroxylase (TH) activity, revealing a negative feedback loop with Ser40 phosphorylation both in vitro and ex vivo, which directly influences on l-3,4-dihydroxyphenylalanine (l-DOPA) production. Maternal Zika virus (ZIKV) infection during pregnancy has been associated with severe intrauterine growth restriction (IUGR), placental damage, metabolism disturbances, and newborn neurological abnormalities. Here, we investigated the impact of maternal ZIKV infection on placental nutrient transporters and nutrient‐sensitive pathways. Immunocompetent (C57BL/6) mice were injected with Low (103 PFU‐ZIKVPE243) or High (5 × 107 PFU‐ZIKVPE243) ZIKV titers at gestational day (GD) 12.5, and tissue was collected at GD18.5 (term). Fetal–placental growth was impaired in male fetuses, which exhibited higher placental expression of the ZIKV infective marker, eukaryotic translation initiation factor 2 (eIF2α), but lower levels of phospho‐eIF2α. There were no differences in fetal–placental growth in female fetuses, which exhibited no significant alterations in placental ZIKV infective markers. Furthermore, ZIKV promoted increased expression of glucose transporter type 1 (Slc2a1/Glut1) and decreased levels of glucose‐6‐phosphate in female placentae, with no differences in amino acid transport potential. In contrast, ZIKV did not impact glucose transporters in male placentae but downregulated sodium‐coupled neutral amino acid 2 (Snat2) transporter expression. We also observed sex‐dependent differences in the hexosamine biosynthesis pathway (HBP) and O‐GlcNAcylation in ZIKV‐infected pregnancies, showing that ZIKV can disturb placental nutrient sensing. Our findings highlight molecular alterations in the placenta caused by maternal ZIKV infection, shedding light on nutrient transport, sensing, and availability. Our results also suggest that female and male placentae employ distinct coping mechanisms in response to ZIKV‐induced metabolic changes, providing insights into therapeutic approaches for congenital Zika syndrome. - Cuts to postgraduate funding threaten Brazilian science — again

Metal encapsulation delivers a straightforward strategy to improve miscellaneous nanoparticle properties and qualifies the resulting nanocomposite for exceptional application, including bioimaging, drug release, and theranostic development. Besides crucial applications, investigations associated with the nanocomposite impact on the biological media are highly relevant from a pharmacological viewpoint. Such studies can be conducted by exploring nanocomposite attributes and all aspects of their interaction with proteins existing in biofluids. Based on these aspects, the present work examines manganese-encapsulated carbonaceous nanocomposite (MnCQD) and their interaction with plasma proteins. On one side, the obtained nanocomposite has almost spherical shapes (≈12 nm in size), an appropriate composition and interesting optical properties for bioimaging applications. On another side, MnCQD quenches the fluorescence of two plasma proteins (BSA and HTF) following a static mechanism, confirming the formation of the MnCQD-BSA and MnCQD-HTF complexes. Although hydrophobic forces guide the stability of both formed complexes, MnCQD binds preferentially to BSA compared to HTF, with affinity constants differing by almost an order of magnitude. Furthermore, HTF and BSA underwent modifications in their secondary structure provoked due to contact with the nanocomposite, which also presented neglectable opsonization levels when exposed to appropriate biological media. Glioblastoma (GB) is the most aggressive primary malignant brain tumor and is associated with short survival. O-GlcNAcylation is an intracellular glycosylation that regulates protein function, enzymatic activity, protein stability, and subcellular localization. Aberrant O-GlcNAcylation is related to the tumorigenesis of different tumors, and mounting evidence supports O-GlcNAc transferase (OGT) as a potential therapeutic target. Here, we used two human GB cell lines alongside primary human astrocytes as a non-tumoral control to investigate the role of O-GlcNAcylation in cell proliferation, cell cycle, autophagy, and cell death. We observed that hyper O-GlcNAcylation promoted increased cellular proliferation, independent of alterations in the cell cycle, through the activation of autophagy. On the other hand, hypo O-GlcNAcylation inhibited autophagy, promoted cell death by apoptosis, and reduced cell proliferation. In addition, the decrease in O-GlcNAcylation sensitized GB cells to the chemotherapeutic temozolomide (TMZ) without affecting human astrocytes. Combined, these results indicated a role for O-GlcNAcylation in governing cell proliferation, autophagy, cell death, and TMZ response, thereby indicating possible therapeutic implications for treating GB. Glutamine:fructose-6-phosphate amidotransferase (GFAT) is the rate-limiting enzyme of the hexosamine biosynthetic pathway (HBP), an important route for de novo synthesis of amino sugars, which are key components of prokaryotic cell walls, chitin, and complex eukaryotic glycoconjugates. GFAT also plays a major role in several pathological processes, including cancer and diabetes. It has been 60 years since GFAT was first characterized. During this time, the knowledge about the enzyme’s mechanisms and biological relevance has increased considerably. We take the anniversary of GFAT’s discovery as an opportunity to discuss the role of GFAT in both health and disease and explore its biotechnological potential as a target for antimicrobial and anticancer chemotherapy. - Protein glycosylation in cancer
