Revolutionizing HPV Detection: Unveiling a Novel Electrochemical Immunosensor for Ultra-Sensitive HPV16 E7 Oncoprotein Detection Based on Gold-Graphitic Carbon Nitride Nanocomposite Hiranmoy Kotal, Thangapandi Kalyani, Arumugam Sangili, Ranajit Kumar Mandal, Suvankar Ghorai, Saikat Kumar Jana Journal of the Electrochemical Society, 2024 Cervical cancer (CC), a significant contributor to the mortality of females globally, manifests an annual incidence of 500,000 cases and is predominantly linked to the human papillomavirus (HPV) infection. The pressing demand for affordable and user-friendly assays for CC detection is highlighted by the limited availability and dependability of diagnostic screening tools, especially in developing countries. In response to the critical need for enhanced cervical cancer diagnostics, we present an innovative electrochemical immunosensor designed for the selective detection of the HPV 16-E7 oncoprotein. The immunosensing platform is constructed from a gold-graphitic carbon nitride (Au-g-C3N4) nanocomposite, with antibodies immobilized through a physical adsorption process onto the sensing platform. Employing Square Wave Voltammetry (SWV), we systematically evaluated the antigen-antibody interaction, achieving ultrasensitive detection of E7 with ferric/ferrocyanide. The linear detection range of the proposed sensor for E7 antigen is 0.01 fg ml−1 to 1 ng ml−1, assuming ideal conditions and an impressive Limit of Detection (LOD) of 0.0065 fg ml−1. The accuracy of the proposed immunosensor was effectively evaluated in contrast to Enzyme linked immunosorbent assay (ELISA). Finally, the proposed immunosensor exhibited favourable selectivity, sensitivity, and reproducibility upon introduction to clinical samples, thereby validating its potential suitability for point-of-care testing aimed at the timely detection of CC.
Sustainable Synthesis of Ruthenium-Palladium-Based Nanonet Assembly for Efficient Reduction of 4-Nitrophenol and Nitrofurantoin Pitchaimani Veerakumar, Arumugam Sangili, Shen-Ming Chen, Naveen Karuppusamy ACS Applied Nano Materials, 2023 Polyol techniques have recently emerged as viable soft chemical approaches for the production of ultrafine metal crystals. In this work, we described a simple method of ruthenium nanochains ornamented on an octahedral palladium (RuNCs@Oct-Pd) hybrid nanonet assembly synthesized by using polyvinylpyrrolidone (PVP, stabilizing agent) and 1,2-propanediol (1,2-PD, reducing solvent). The prepared nanonet assembly plays a vital role for reduction of toxic nitro substances such as (4-nitrophenol, 4-NP) and hazardous antimicrobial agents (nitrofurantoin, NFT). The morphology of the nanonet assembly was tested by XRD, SEM, FE-TEM, XPS, and UV–vis spectral techniques. The RuNCs@Oct-Pd nanonet assembly demonstrates enhanced catalytic reduction of 4-NP and NFT using NaBH 4 (reductant) at ambient temperature. The RuNCs@Oct-Pd hybrid nanonet assembly exhibited a higher rate constant ( k app = 0.4332 s –1 ) and turnover frequency (TOF = 5.0 × 10 –2 s –1 ) than commercial catalysts like Ru/C and Pd/C because of substantial catalytic active sites, high surface-to-volume ratio, rapid surface-hydrogen transfer, and superior stability. Furthermore, the hybrid catalyst in this combination showed outstanding durability and reusability, making it suitable for real-world applications.
Octahedral Pt-Ni Alloy Nanoparticles Decorated on 3D Interconnected Porous Carbon Nanosheets for Voltammetric Determination of Dihydroxybenzene Isomers Pitchaimani Veerakumar, Arumugam Sangili, Shen-Ming Chen, Venkatachalam Vinothkumar, Tae Hyun Kim ACS Applied Nano Materials, 2023 Ultrathin three-dimensional (3D) interconnected porous carbon nanosheets (PCNs) derived from Moringa oleifera barks (commonly known as the drumstick tree) with electrochemical (EC) sensor performance was employed as the carbon precursor. In this study, we described a simple one-step pyrolysis technique to generate hierarchical PCN using a high-temperature pyrolysis and KOH activation procedure. Using the microwave-solvothermal approach, platinum–nickel bimetallic octahedral nanoparticles were decorated on ultrathin 3D PCN (referred to as Pt–Ni@PCN). The morphological, structural, and EC properties of the Pt–Ni@PCN nanocomposite were well characterized. Using cyclic voltammetry and linear sweep voltammetry, the dihydroxybenzene (DHB) isomers hydroquinone, catechol, and resorcinol were identified individually as well as simultaneously. The screen-printed carbon electrode-modified nanocomposite [Pt–Ni@PCN/screen-printed carbon electrode (SPCE)] served as an outstanding electron (e–)-transfer mediator for DHB isomer oxidation, yielding limits of detection of 0.0093, 0.0263, and 0.0529 μM, respectively. Also, the Pt–Ni@PCN/SPCE sensor has exceptional selectivity, long-term durability, reproducibility, repeatability, and anti-interference. Furthermore, the proposed sensor can be exploited to detect simultaneously environmental pollutants in river, lake, and tap water samples with good recovery rates.
Ultra-sensitive label-free detection of haptoglobin using Au-rGO decorated electrochemical sensing platform: Towards endometriosis diagnostic application Thangapandi Kalyani, Arumugam Sangili, Hiranmoy Kotal, Ajeet Kaushik, Koel Chaudhury, Saikat Kumar Jana Biosensors and Bioelectronics X, 2023 In this research, for the first time, one-step electrodeposition of simultaneously gold nanoparticles (AuNPs) and reduced graphene oxide (rGO) on the surface of a glassy carbon electrode (GCE) was used as a sensing platform of highly sensitive and label-free immunosensing for the rapid voltammetric analysis for haptoglobin (HP) at ultra-trace levels. 11-mecaptoundecanoic acid (11-MUA) was self-assembled onto Au substrate, which was coupled to the antibody (ab-HP) via ethyl-N’-(3-dimethyl aminopropyl) carbodiimide hydrochloride–N-hydroxysuccinimide (EDC-NHS) coupling. The interaction between the antigen (HP) and antibody (ab-HP) is measured by square wave voltammetry in [Fe (CN)6]3−/4− solution. The proposed immunosensor exhibits a concentration that ranges from 10 fg mL−1 to 1000 ng mL−1, and 3.47 fg mL−1 of low detection limit (LOD). Moreover, an immunosensor has high sensitivity, selectivity, reproducibility, stability, and reusability. Ultimately, this system was successfully applied for the detection of HP in different positive and negative serum samples (positive: n = 40, negative: n = 10), whereas negligible change was observed in control samples and was compared with commercial ELISA kits. The specificity and sensitivity of the assay were 100% and 98%, respectively. The coincidence rate using the HP ELISA kit was 95.30% and 92.20%. The result demonstrated that the sensing platform could be used for clinical diagnosis of HP in clinical laboratories.
