Determining the Role of KRAS in Patients with Different Demographic Profiles Diagnosed with Diabetes Mellitus: A Case-Control Study Noora A. Hadi, Rehab S. Ramadhan, Khalid Suhail A. AlAzzawi, Rebah N. Algafari, Sura S. Talib, Rawaa A. Khalaf Obm Genetics, 2026 Diabetes mellitus (DM) is widely spread among populations. About 3 of 10 people show the symptoms of this disease. Many factors may cause this illness in both types (type 1 and type 2 DM), mainly attributed to insufficient insulin production in type 1 DM or developing insulin resistance in type 2 DM. Genes that control specific biochemical pathways involving glucose metabolism can interfere with the manifestation of this disease when they undergo genetic changes like KRAS. Investigating the role of KRAS in DM. About 124 patients with both types of DM and of different genders and ages participated in this study. In addition to demographic characteristics, biochemical and diabetic status were determined for all participants. DNA was isolated from all patients, and the KRAS gene was amplified using specific primers designed for this purpose and thoroughly analyzed to determine any genetic change within its sequence. The results showed the spread of DM, especially type 2, among undereducated participants, and a significant increase in fasting blood sugar, uric acid, and SGOUT. Molecular analysis of the KRAS gene identified two repeated single-nucleotide variations (SNVs): rs1475732807 and rs1306089842. Both SNVs affected the 3’ UTR of the KRAS gene, causing an alteration in gene expression, which consequently affected the severity of DM. Diabetes mellitus is widely spread among different societies, especially in undereducated groups, due to unhealthy diets or genetic factors. Some genes, like KRAS, can interfere with the manifestation of DM, especially related to glucose metabolism. SNVs in the 3’ UTR of KRAS may dramatically boost the symptoms in some patients.
The Possible Role of Progesterone Receptors in Prostate Cancer Incidences in the Iraqi Population Jaleel Ibrahim Asaad, Khalid S. A. Alazzawi, Sara S. Rasheed, Rebah N. Algafari, Rehab S. Ramadhan, Marwah Amer Qamandar, Sura S. Talib, Rawnaq Z. Fadhil Frontiers in Bioscience Scholar, 2024 Background: Prostate cancer (PCa) is one of the leading diseases causing mortality. It comes in the third rank of common cancer types. It is considered extremely a complicated cancer type since it occurs in highly steroid-responsive and dependent tissues. Many factors are considered to play an important role in the disease progression of PCa, with some functioning at the molecular level. Methodology: After applying the exclusion criteria, 200 patients who underwent proctectomy were included in this study. Following receiving patient consent, blood samples were withdrawn from patients, DNA was extracted, and precise polymerase chain reaction (PCR) amplification was conducted using specifically designed primers. The resulting amplicons were sequenced and analyzed. Results: The progesterone receptor B (PGRB) DNA from patients showed four distinctive single-nucleotide polymorphisms (SNPs) at sites 11:101128812, 11:101128924, 11:101128949, and 11:101128986, which altered the amino acid sequences to Y>N, A>D, T>I, and C>R, respectively, compared to control. These SNPs resided in sensitive sites that either affected the control elements or promoted alterations in the protein configuration. This DNA change diminished the PGR gene function and promoted an imbalance in the encoded PGR protein structure and expression. Conclusions: Many factors may play a role in PCa manifestation, with steroids and progesterone initially noted as factors. Many studies have dealt with the hormonal effect on PCa; however, few have ultimately determined the molecular impact on disease progression. The presence of pathogenic SNPs in the enhancing region of the gene may impact the expression level of PGR. High or low expression levels may negatively affect gene function, which can be considered a reliable factor in prostate tumorigenesis.
In Vitro Effects of Acetaminophen Drug(Paracetamol) on Microscopic Human Sperm Parameters Rana R Al. Saadi, Ban Thabit Al-Ani, Khalid S. A. Alazzawi Aip Conference Proceedings, 2022 Paracetamol, also recognized as acetaminophen (CH3CONH.C6H4), is a drug used to relief pain and fever. It is usually used to relief mild to moderate pain . Forty semen samples were taken from forty infertile patients. After semen analysis assessment, The samples were classified into 2 aliquots group, Pre - activation and Post-Activation group. The Post-Activation group was divided into three subgroups: First subgroup G1 (control group) containing semen sample with in vitro Flashing activation medium. Second subgroup G2 containing semen sample with in vitro Flashing activation medium supplemented by Paracetamol drug powder (0.0226 g/l). Third subgroup G3 containing semen sample with in vitro Flashing activation medium supplemented with Paracetamol drug powder (0.0453 g/l). Direct swim up and centrifugation technique was used for sperm activation in vitro . Sperm parameters were assessed and data were analyzed statistically pre- and post-activation in G1,G2 and G3. The results showed a significant increase in the percentage of sperm motility (%) in G1 and G2 groups (P<0.05) in post-activation when compared with pre-activation. However, there was no significant difference in the post-activation sperm total motility (%) between the G1 and G2 groups. Also there is significant difference (P<0.05) of normal sperm morphology observed between Pre- and Post-Activation using Flashing medium supplemented with Paracetamol drug and there is non-significantly different (P>0.05) of post activation of subgroup G1 and G2 and G3. There were significant different (P<0.05) sperm concentration seen between Pre- and Post-activation using Flashing medium supplemented with Paracetamol Drug while there is non-significantly different (P>0.05) of post activation groups compare with each other G1,G2 and G3. The results of this clinical research indicated paracetamol as antipyretic, analgesic drug have no significant (in vitro) effect on sperm motility, concentration, morphology in post activated semen samples. So this drug not preferred to use for increase IVF technique successful rate.
Benefit effect of ethanolic extract of Bay leaves (Laura nobilis) on blood sugar level in adult diabetic rats induced by alloxan monohydrate Salah M.M. Al Chalabi, Duha Mysire Majeed, Ahmed Abdulmunem Jasim, Khalid Suhail Al-Azzawi Annals of Tropical Medicine and Public Health, 2020 In the present study, the effect of Bay leave ethanolic extract on blood glucose, insulin level, lipid profile , liver and kidney function and body weight in type 2 diabetic rats were investigated. Forty albino male rats(250-300) grams were divided into four groups (10 rats for each), first group treated with distilled water and served as control, second group treated with 200 mg\\kg of Laura nobilis alcoholic extract, third group served as positive diabetic and the fourth group was served as diabetic and treated with 200 mg\\kg b. w of alcoholic extract of Laurus nobilis, all groups were treated orally, the following results were evaluated in diabetic rats, fasting blood glucose, triglyceride (TG), total cholesterol (TC), low density lipoprotein (LDL), very low density lipoprotein (VLDL),alanine aminotransferase (ALT),aspartate aminotransferase (AST) ,alkaline phosphatase (ALP) , B. urea and s. creatinine levels were significantly increased while high density lipoprotein (HDL) and body weight significantly decreased as compared to control rats, after 30 days of treating by200 mg\\kg b.w alcoholic extract of Bay leave there was significant decrease in fasting blood glucose and significant increase in fasting insulin level, also there was significant decrease in TG, TC, LDL, VLDL, ALT, AST, ALP, B. urea and s. creatinine level, while HDL and body weight increased in diabetic groups compared to control not treated groups. The effect of mechanical vibration on human sperm activity in vitro Ghassan Thabet Saeed, Khalid Suhail A. Al-Azzawi, Hydar Saadi Hassan Al-Wasti Biomedical and Pharmacology Journal, 2018 It is well known that sperm is a unique cell in that it has a function to be done by itself outside the body and this function is essential for species' continuity thus sperm by its power and intact structure has to reach the ova and perform the fertilization and this journey is affected by the chemical and physical factors that might increase or decrease its ability to move or fertilize or even to survive. The aim of this study is to find the effect of vibration that is a vigorous movement with high frequency for 20min on whole seminal fluid samples as an external physical factor. 40 fresh seminal fluid samples were selected. 1ml of each semen samples was placed in the bottom of conical tube; the tube was exposure to vibration waves by using a special shaker designed for this purpose for 20 min. This shaker consist of a M540 DC motor equipped with PWM controller to control the rotational speed from 5-2400 rpm. Semen analysis was done before and after subject vibration. A significant increase (P<0.05) was found in percentage of sperm active directed motility (grade A) with a non-significant increase in sluggish motility and a non-significant decrease in percentage of immotile sperms percentage. No significant changes were founded regarded sperm morphology and count. It was concluded that vibrating seminal sample for 20min increases the overall sperms activity with significant increase in percentage of highly active directed sperms.
