  • Disposable pipette extraction phase based on styrene-divinylbenzene/ pernigraniline composite, applied for dexamethasone determination in synovial fluid by HPLC with UV detector

    A reliable method using disposable pipette extraction (DPX) based on composite of pernigraniline and styrene–divinylbenzene (Sty–DVB) copolymer was applied to the analysis of dexamethasone in synovial fluid using high-performance liquid chromatography and ultraviolet (UV) detector (HPLC–UV). DPX variables, namely, number of draw/eject cycles for extraction or desorption, sample pH, volume, and desorption solvent, were optimized to establish the best sorption equilibrium and analysis time. The highest extraction efficiency value was obtained with 50 μL of synovial fluid mixed with 1950 μL of water, in five cycles of 300 μL of sampling, followed by liquid desorption of the drug with 300 μL of methanol in three cycles. The developed method demonstrated a linear response over the range from 10 to 100 ng/mL, with R2 = 0.993. The limit of quantification (LOQ) was 10 ng/mL. Based on the validation results, the proposed method can be a useful tool to detect dexamethasone levels in synovial fluid.

  • Factorial design for evaluation of reagent concentrations on silver nanoparticles stability

    Nanomaterials, especially silver nanoparticles (AgNps) offer unique optical, chemical and physical properties when compared to properties of bulk materials due to their plasmonic ressonance band. Despite AgNps synthesis by chemical reduction is not new, their stability is a crucial factor for a good applicability. AgNps were characterized by UV-vis spectroscopy. Different reports found in literature related to concentrations of used reagents in nanoparticles synthesis are scares when one take into account the high control that is required to avoid nanoparticles aggregation. This study aims to evaluate the concentrations of the reducing agent – NaBH4 (factor a), stabilizing agent – PVA (factor b) and AgNO3 (factor c) through a factorial design 23 with three center points of face centered to evaluate the influence of reagents and their concentrations on AgNps stability. The interaction between factors “a” and “c” showed a positive significant value when concentrations are modified from level (-) to level (+) of these two variables. It can be observed an increase of plasmonic band absorption peaks of 18.4% indicating synergism between variables a and c. However, one can notice a decrease of 3.91% of peak’s incidence when factor “b” concentrations are increased. It can be inferred that low PVA concentrations (≤ 0.5%) assist the AgNps stability and above this value PVA acts as a chemical inhibitor. Analysis of variance indicated a coded quadratic model of second order. Plots of response surface showed that the ideal synthesis of AgNps happens on the ratio NaBH4 (mmol) : PVA (%) : AgNO3 (mmol) of 4:1:2.

  • Nanostructured sensors containing immobilized nuclear receptors for thyroid hormone detection

    Thyroid hormone receptors (TRs) are members of the nuclear receptors (NRs) superfamily, being encoded by two genes: TRa and TRbeta. In this paper, the ligand-binding domain (LBD) of the TRbeta1 isoform was immobilized on the surface of nanostructured electrodes for TR detection. The platforms containing TRbeta1-LBD were applied to the detection of specific ligand agonists, including the natural hormones T3 (triiodothyronine) and T4 (thyroxine), and the synthetic agonists TRIAC (3,5,3'-triiodothyroacetic acid) and GC-1 [3,5-dimethyl-4-(4'-hydroxy-3'-isopropylbenzyl phenoxy) acetic acid]. Detection was performed via impedance spectroscopy. The biosensors were capable of distinguishing between the thyroid hormones T3 and T4, and/or the analogues TRIAC and GC-1 at concentrations as low as 50 nM. The detection and separation of thyroid hormones and analogue ligands by impedance techniques represents an innovative tool in the field of nanomedicine because it allows the design of inexpensive devices for the rapid and real-time detection of distinct ligand/receptor systems.
