    The endocannabinoid system (ECS) is a complex signaling network essential for regulating various physiological processes in the body. Selective cannabinoid receptor ligands have been developed to modulate specific ECS signaling pathways, offering potential therapeutic benefits. These ligands, with high selectivity and affinity for cannabinoid receptors, demonstrate potential in managing diverse medical conditions. Standardizing dosing is crucial to ensure reliable therapeutic effects, as cannabinoids may exhibit biphasic effects. Combination strategies involving both CB1 and CB2 receptor modulation show promise in managing complex conditions, including chronic pain, autoimmune disorders, and neurodegenerative diseases.

    This chapter contains, in brief about the aging of skin and the molecular mechanisms involved therein. The symptoms of wrinkles, fine lines, pigmentation, and saggy and dry skin manifest in skin aging. Reactive oxygen species, reactive nitrogen species, Metalloproteases (MMPs), tyrosinase enzymes, and SASPs play a significant role. With the increasing awareness of people towards skin health, the demand for active ingredients to combat aging increases. Being light, non-toxic, stable, and moisturizing, peptides have been proven extremely beneficial in improving skin conditions. These peptides act via several mechanisms when applied locally or taken orally. Signal peptides, Carrier peptides, Neurotransmitter-inhibiting peptides, and Enzyme-inhibiting peptides act via different mechanisms. In the present chapter, a detailed review of the different types of peptides, their mechanisms, and their action to prevent skin aging have been described. These peptides help to improve skin damage, improves skin elasticity, softness and firmness. It also helps in whitening, rejuvenation, repair and thickening of skin. Peptides, being economic, have opened up a new era in cosmeceuticals. Current markets are flooded with peptide-containing anti-aging products with proven clinical efficacy.
  • DESIGNING A SUSTAINED-RELEASE SOLID ORAL FORMULATION FOR OVERACTIVE BLADDER TREATMENT: A QUALITY BY DESIGN APPROACH

    Background: Mirabegron, a first-in-class β3-adrenergic agonist used for managing overactive bladder (OAB), is well-documented in the literature. However, its low oral permeability results in poor bioavailability, limiting patient compliance and tolerability. To address this, the present study focuses on developing a sustained-release (SR) tablet of mirabegron with enhanced oral effectiveness. In this research, mirabegron was combined with polyethylene oxide to improve permeability and formulated as an HPC-loaded SR tablet, promising improved bioavailability and anti-OAB efficacy. Methods: Tablet design and optimization were carried out using the Box-Behnken Design (Design Expert® 12 software) to refine formulation parameters. The study aimed to create a commercially viable SR tablet with improved intestinal permeability, bioavailability, and clinical acceptance. Results: The optimized formulation showed a 34.8% increase in bioavailability compared to the marketed tablet. In vivo pharmacokinetic studies demonstrated a 31.77% increase in plasma concentration over the marketed formulation. Conclusion: The developed formulation is safe and effective, offering improved therapeutic potential for treating overactive bladder. This work represents a significant advancement in OAB management and highlights the commercial viability of the emerging mirabegron SR tablet in meeting current therapeutic needs.
  • Revolutionizing Ophthalmic Care: A Review of Ocular Hydrogels from Pathologies to Therapeutic Applications

    PURPOSE This comprehensive review is designed to elucidate the transformative role and multifaceted applications of ocular hydrogels in contemporary ophthalmic therapeutic strategies, with a particular emphasis on their capability to revolutionize drug delivery mechanisms and optimize patient outcomes. METHODS A systematic and structured methodology is employed, initiating with a succinct exploration of prevalent ocular pathologies and delineating the corresponding therapeutic agents. This serves as a precursor for an extensive examination of the diverse methodologies and fabrication techniques integral to the design, development, and application of hydrogels specifically tailored for ophthalmic pharmaceutical delivery. The review further scrutinizes the pivotal manufacturing processes that significantly influence hydrogel efficacy and delves into an analysis of the current spectrum of hydrogel-centric ocular formulations. RESULTS The review yields illuminating insights into the escalating prominence of ocular hydrogels within the medical community, substantiated by a plethora of ongoing clinical investigations. It reveals the dynamic and perpetually evolving nature of hydrogel research and underscores the extensive applicability and intricate progression of transposing biologics-loaded hydrogels from theoretical frameworks to practical clinical applications. CONCLUSIONS This review accentuates the immense potential and promising future of ocular hydrogels in the realm of ophthalmic care. It not only serves as a comprehensive guide but also as a catalyst for recognizing the transformative potential of hydrogels in augmenting drug delivery mechanisms and enhancing patient outcomes. Furthermore, it draws attention to the inherent challenges and considerations that necessitate careful navigation by researchers and clinicians in this progressive field.

    Most cancers have become immune to normal cancer therapy, like chemotherapy and radiation. Therefore, exploring more effective and economical treatment options is important. Plants and herbs contain substances called phytochemicals, which have biological effects. Many phytochemicals having antioxidant and anticancer properties have been studied previously. There is increasing evidence that phytochemicals' anti-carcinogenic benefits originate from their ability to inhibit oxidation, inflammation, cell proliferation, and angiogenesis. These phytochemicals inhibit the spread of cancer by controlling the cell cycle and other molecular processes, such as metastasis. Along with therapeutic potential, other advantages, like their abundance, greater tolerability, and economic use, increase their utility in cancer therapeutics. In recent years, a number of scientists have examined lycophytes and ferns for their potential medicinal and phytochemical properties. This analysis emphasizes the significance of chemicals obtained from ferns and their derivatives in therapeutics. The authors discuss the pteridophyte's anti-cancer properties and other medical uses in this article. This information may help researchers in further research related to the most promising anticancer phytochemicals and their possibility as alternative drugs against cancer.

    Biopolymers as Therapeutic Adjuvants Innovations and Advancement, 2025
    To foster a green environment, considerable efforts have been made to replace synthetic polymers with biodegradable materials, such as biopolymers, particularly for the development of green drug delivery systems. Biopolymers are a prominent class of functional materials with high-value applications, generated either by biological systems or derived from biological sources. Natural sources of biopolymers include plants, animals, microorganisms, and agricultural wastes. Biopolymers exhibit excellent properties, including flexibility, tensile strength, stability, reusability, and many more. Biopolymers are composed of repetitive monomers bound covalently via polymerization reaction or enzyme-catalyzed assemblies of monomeric units that occur in the biosynthetic pathway within biological systems. Biopolymers can be classified based on their source, chemical composition, functional properties, degradability, type of charges, and other factors. The extraction of biopolymers involves a range of chemical and enzymatic processes that vary specifically for each biopolymer. Some of the extraction methods include the use of coagulating agents, hydrolysis, alkali and acid treatments, bleaching, deproteination, and demineralization, among others. Following extraction, purification, and often modification, biopolymers are prepared for potential applications. Due to their renewability, abundance, biodegradability, and unique properties, such as higher absorption capabilities and ease of functionalization, biopolymers have been explored for various industrial applications. This chapter examines the sources, chemistry, and extraction procedures for several important biopolymers, including polyhydroxyalkanoates, polylactic acid, chitosan, alginate, polyesteramide, starch, gelatin, polyglycolic acid, and pectin, as well as their biomedical applications.

    Women S Health A Comprehensive Guide to Common Health Issues in Women, 2024
    Unwanted pregnancies and unmet contraceptive needs are significant global public health issues. Increased contraceptive usage in poorer nations over the past 20 years has reduced maternal fatalities by 40% through a decline in unplanned births. Starting contraception at any stage of the menstrual cycle is generally safe, promoting health and reproductive autonomy. Meeting the unmet demand for contraception could prevent an additional 30% of maternal fatalities. Contemporary contraceptives offer advantages for Women's health, surpassing the disadvantages. Postpartum contraception timing and substance are being modified to address issues like unwanted pregnancies and rapid, recurrent pregnancies. Progestin-only contraceptives can be initiated immediately after childbirth, and any suitable method is safe following abortion or early pregnancy loss. Delaying contraception based on periods or appointments puts individuals at needless risk. To encourage contraceptive use, clinicians should inform patients about common side effects, provide comprehensive information on available options, respect patient preferences, and remove barriers to access. The adolescent prenatal period is a pressing concern, requiring changes in laws and customs to state young individuals' requirements. Limited access to family planning (FP) resources and services affects adolescents, while cultural traditions and legislation hinder their access to medical care. Abortions among young women have increased and become more physically distressing. Addressing these challenges necessitates comprehensive reproductive healthcare services, education, policy changes, and improved access to contraception. By prioritizing these efforts, we can reduce maternal fatalities, empower individuals to make informed reproductive choices and support the well-being of women and adolescents globally.
