The world has recently turned to find alternatives to plant growth regulators, especially materials used in plant tissue culture, as they are the main pillar in establishing tissue cultures and participate in all stages of agriculture. Plant extracts from (seeds or leaves) are most important that can encourage growth, as they contain many active compounds that may compensate for hormones and increase the plant's resistance to drought. Moringa extract contains phenols, ascorbic acid, in addition to growth-promoting hormones (auxins, gibberellins and cytokinins), and abscisic acid, and the activities of the antioxidant enzymes (catalase, peroxidase, and superoxide dismutase). In the research, barley embryos were grown in the Murashige and Skoog (MS) culture media containing of Moringa leaf extract with (0, 1.5, 3 and 4.5 mL L-1), versus kinetin (Kin) treatments at 0, 1, 2 and 3 mg L-1 and indole acetic acid (IAA) at 0.5 mg L-1 then, plantlets that achieved highest average number of shoots, total chlorophyll and dry weight were transferred to the culture media containing Poly Ethelene Glycol (PEG) at 0, 2, 4 and 6 g L-1 and the work was performed from a fully Complete Randomized Design (CRD), the sample size on which the analysis was conducted was 160 samples resulting from the use of two varieties of barley, 4 concentrations of Moringa extract, 4 concentrations of Kin, and 5 replications. The results estimated that the highest average number of the vegetative shoots formed was 16.8 shoots plant-1, the total amount of chlorophyll was 323.4 mg g-1, and the dry weight of vegetative shoots was 88.9 mg in concentration 3 of Moringa and Kin extract in the Buhuth244 variety. The average plant relative growth rate was 49.2 mg day-1, and the average plant growth rate was 56.1 mg day-1 in Buhuth244, and 3 mL L-1 of Moringa extract and 4 of PEG. The examination, diagnosis, and estimation of the active compounds in the Moringa leaf methanolic extract was identified by Gas Chromatography with Mass Spectrometer unit (GC-MS). These extracts can serve as useful antioxidant materials. This study has concluded that Moringa extract can be used as an alternative to synthetic plant hormones due to its effect on improving vegetative growth characteristics.
THE EFFECT OF HARMEL SEED EXTRACT ON THE VEGETATIVE GROWTH CHARACTERISTICS AND DROUGHT TOLERANCE OF SUNFLOWER IN VITRO , SJS Baday, MT Aljubori, MI Salih African Journal of Food Agriculture Nutrition and Development, 2025 The world has recently become focused on finding alternatives to plant growth regulators, particularly those used in plant tissue culture, as they are the cornerstone of establishing tissue cultures and are involved in all stages of agriculture. Plant extracts are one of the most important factors that encourage growth, as they contain many active compounds that can replace hormones and increase plant resistance to drought. This study aimed to use harmala extract as an alternative to artificial hormones and to induce genetic variations that would increase the ability of plants to tolerance stress. In this study, sunflower embryos were grown in media known as Murashige and Skoog (MS) culture media that containing (0, 4, 6 and 8) ml L-1 of alcoholic extract of Peganum harmel seeds, as compared to benzyl adenine (BA) treatments of (0, 1, 2, and 3) mg L-1 and indole acetic acid (IAA) of 0.5 mg L-1. The plantlet that recorded the highest average number of vegetative shoots and dry weight were transferred to a culture medium containing mannitol compound from (0, 5, 10, and 15) g L-1. The results experimentally showed that the actual highest average number of vegetative shoots formed was 22.6 shoots plant -1, and the total amount of chlorophyll was 983.9 mg g-1, and the dry weight of vegetative shoots was 303.4 mg at 6 ml L-1 of harmel seed extract and 2 mg L-1 of BA in the Elf variety. The relative average growth was 66.2 mg day-1, while the plant growth average was 73.8 mg day-1 and the stress tolerance index was 2.321 in the Elf variety and the 6 ml g-1 of harmel seed extract and 10 g L-1 of mannitol. The polymerase chain reaction-rapid amplification polymerase DNA (PCR-RAPD) revealed a difference in the number of bands and their molecular weights for the harmel extract treatments compared to the comparison treatment. The Importance of Cryoprotective Agents (CPAS) in the Cryopreservation Process for the Preservation of Plant Resources Mayada T. Al-Jubori, Rasha Raad Mohammed Iop Conference Series Earth and Environmental Science, 2024 The process of vitrification involves the freezing of plant genetic components. It is based on the fact that water can be rapidly cooled to a glassy state by increasing its viscosity to extremely high values without causing intracellular ice crystallization. The genetic material of plants can undergo deleterious changes in structure and function during the cryopreservation process. It is important to remember that cryodamage can occur at any time after ice freezing, not just during the freezing process. Cryopreservation and thawing have been associated with potentially greater changes in DNA integrity. Due to the increased rate of lipid peroxidation and suppression of the antioxidant enzyme defense mechanism, cryopreservation causes oxidative stress. Cryopreservation is particularly damaging to RNA and DNA. The development of cryopreservation technology is due to the discovery of chemicals with preservative properties and protection against freezing damage. These chemicals must be applied to the culture medium for the culture to be preserved in order to prevent the formation of large ice crystals in the plant cells; the sharp edges of these crystals damage the cell walls and membranes and kill the cells.
Effect of Foliar Spray of Zinc and Irrigating with Smoking-Water on Growth of Citrus Rootstock C35 Ausama Y. Salih, Mayada Tariq Al-Jeboory Iop Conference Series Earth and Environmental Science, 2023 This study was conducted in fruit production lathe house of the Department Of Horticulture And Landscape Gardening, in the station (B), College Of Agricultural Engineering Sciences, University Of Baghdad, Al-Jaadria for 8 months, began from 1/3/2019 to 1/9/2019 to investigate the responses of C35 Citrus rootstock to influence of foliar spraying of zinc and irrigation with smoking-water at 2 years old saplings. the study included two factors, first factor was three concentrations of Zinc Element Z0(0 mg.l-1), Z1(50 mg.l-1) and Z2(75 mg.l-1) that sprays at leave. The second factor was watering saplings with smoke-water in three concentrations S0(0 vol.vol-1), S1(0.1 vol.vol-1) and (0.2vol.vol-1), and their interaction. The experiment was factorial in RCBD design, Treatments were replicated 3 times, number of saplings were 54. Results showed that zinc at 75 mg.l-1 and smoke-water at 0.2vol.vol-1 Significantly gave the highest vegetative, root and chemical content in the leaves respectively. the lowest value of these parameters was found in control treatment.
OPTIMIZING MEDIA STERILIZATION VIA CHLORINE DIOXIDE AND AUTOCLAVING OF PAULOWNI MICROPROPAGATION M. T. Al-Jubori, F. M. K. AL-Dabbagh, E. W. Al-Ani Iraqi Journal of Agricultural Sciences, 2023 This study was aimed to investigat integrated system for in vitro growth of paulownia plants by assessing the efficacy of chlorine dioxide (ClO2) as an alternative to autoclave in sterilizing culture medium. Therefore, this study was devised to compare autoclave sterilization at three different times (5, 10, and 15) minutes and three different concentrations of ClO2 (0, 0.4, 0,8, 1) mg/L. The results showed that, compared with (0.4) mg/L concentration, concentrations of (0.8 and 1) mg/L are more effective at sterilizing the culture medium. ClO2 sterilization improved individual single node growth more than autoclave sterilization. Since ClO2 is non-toxic, it could be used as a safe alternative to autoclave when propagating paulownia in vitro. Culture medium sterilization in the autoclave takes only 5 minutes, compared with the standard 15 minutes. At initiation stage, growing single nodes in the Murashige and Skoog medium (MS) prepared with 0.5 mg/L Benzyl Adenine (BA) resulted in a 100% response rate, while doing the same in the Woody Plant Medium (WPM) resulted in a 20% response rate. The 1 BA + 0 a-Naphthalene Acetic Acid ( NAA) mg/L treatment was effective during vegetative multiplication stage, the highest average number of shoots produced by a plant treated with the mentioned concentration was 6.40 shoot per explant. During the rooting stage, Indole Butyric Acid (IBA) at a concentration of 2 mg/L was more effective than NAA, the typical number of roots produced by with 27.40 root per shoot. After two months in their natural environment, the plants' acclimatization rate was at a perfect 100%.
