  • 8-Oxo-2′-Deoxyguanosine: Oxidative Stress Control

    8-Oxo-2′-deoxyguanosine is a well-known marker of oxidative stress. Research over the past decade suggests that this compound is probably not a byproduct of oxidative DNA damage, but an important bioregulator driving the cellular response to stress. This review collected and analyzed data on the participation of 8-oxo-2′-deoxyguanosine in the processes of mutagenesis, DNA repair, and regulation of gene expression, inflammatory responses, adaptive stress response, apoptosis, and cellular transformation. Particular attention is paid to the potential of 8-oxo-2′-deoxyguanosine as a therapeutic agent for the treatment of inflammatory, autoimmune, degenerative, and oncological diseases, as well as of traumatic and toxic injuries.

    Inorganic polyphosphates (polyP), according to literature data, are involved in the regulatory processes of molecular complex of the Saccharomyces cerevisiae cell wall (CW). The aim of the work was to reveal relationship between polyP, acid phosphatase Pho3p, and the major CW protein, glucanosyltransglycosylase Bgl2p, which is the main glucan-remodelling enzyme with amyloid properties. It has been shown that the yeast cells with deletion of the PHO3 gene contain more high molecular alkali-soluble polyP and are also more resistant to exposure to alkali and manganese ions compared to the wild type strain. This suggests that Pho3p is responsible for hydrolysis of the high molecular polyP on the surface of yeast cells, and these polyP belong to the stress resistance factors. The S. cerevisiae strain with deletion of the BGL2 gene is similar to the Δpho3 strain both in the level of high molecular alkali-soluble polyP and in the increased resistance to alkali and manganese. Comparative analysis of the CW proteins demonstrated correlation between the extractability of the acid phosphatase and Bgl2p, and also revealed a change in the mode of Bgl2p attachment to the CW of the strain lacking Pho3p. It has been suggested that Bgl2p and Pho3p are able to form a metabolon or its parts that connects biogenesis of the main structural polymer of the CW, glucan, and catabolism of an important regulatory polymer, polyphosphates.
  • Pseudo-minimal surfaces of revolution

    This paper is a follow-up to the first author’s series of works about shape modeling of orthotropic elastic material that takes the equilibrium form inside the area with the specified boundaries. M.S. Bukhtyak, in a number of his publications of 20162020, proposed an approach to the model building based on the application of surfaces with a constant ratio of principal curvatures. These surfaces are named pseudo-minimal surfaces. The theorem of existence has been demonstrated and the finitely-element model has been built. The condition distinguishing the class of pseudo-minimal surfaces, as applied to ruled surfaces, is either satisfied identically (trivial subclasses) or is satisfied along a family of lines. The corresponding classes of ruled surfaces have been comprehensively characterized geometrically. A partial differential equation that defines (in the local sense) the class of pseudo-minimal surfaces is very complex for analysis, which makes it relevant to consider approximate solutions. The current paper considers the pseudo-minimal surfaces of revolution. Generation of the approximate solutions is complicated by the tendency of the formal Taylor polynomial to diverge.     New polyanionic modifications of polyamide nucleic acid mimics were obtained. Thymine decamers were synthesized from respective chiral α- and γ-monomers, and their enantiomeric purity was assessed. Here, we present the decamer synthesis, purification and characterization by MALDI-TOF mass spectrometry and an investigation of the hybridization properties of the decamers. We show that the modified γ-S-carboxyethyl-T10 PNA forms a stable triplex with polyadenine DNA.
  • Biological role of 8-oxo-2'-deoxyguanosine

    The review presents currently available data on the biological role of 8-oxo-2’-deoxyguanosine. This compound for a long time has been used successfully as a biomarker of oxidative stress and diseases associated with it. However, in recent years, an increasing number of publications has appeared reporting that biological role of 8-oxo-dG is not only a mutagenic one; it also is involved in the regulation of gene expression, in some processes of DNA repair, in the control of inflammatory and autoimmune reactions, and in the activation of antioxidant systems. Probably, 8-oxo-2’-deoxyguanosine could prospectively be used as a drug.
  • SkBQ - Prooxidant addressed to mitochondria

    Oxidative stress and mitochondrial dysfunction are the key links in the chain of development of pathologies associated with the violation of cellular energy metabolism. Development of mitochondria-addressed compounds highly specific for chemical processes is one of the most promising ways to develop approaches to the treatment of inherited and age-related diseases with mitochondrial etiology. Correlation of structure and chemical activity of the test compounds from a class of lipophilic cations revealed the key role of substituents in the aromatic ring of 1,4-benzoquinones in the manifestation of high antioxidant properties. In this work, it is shown that a synthesized benzoquinone derivative conjugated in position 6 with membrane-penetrating cation of decyltriphenylphosphonium and with substituents at position 2, 3, and 5 (SkBQ) has much lower antioxidant and significantly higher prooxidant activity in comparison with similar derivatives of plasto- and toluquinone SkQ1 and SkQT1 in experiments on isolated mitochondria. At the same time, SkBQ, like SkQ1 and SkQT1, can be reduced by the respiratory chain in the center i of complex III and decrease the mitochondrial membrane potential. In cell cultures of human fibroblasts, it was revealed that SkBQ does not protect cells from apoptosis induced by hydrogen peroxide. Under the same conditions, SkQ1 and SkQT1 exhibit a powerful protective effect. Thus, SkBQ can be seen as a mitochondria-addressed prooxidant. The possibility of using SkBQ as an anticancer drug for the treatment of cancers such as prostate cancer whose cells are sensitive to mitochondrial reactive oxygen species is discussed.
