Raghavendra Rao, working as a Professor in the Department of Pharmacy, KIET School of Pharmacy, at KIET Group of institutions, Delhi-NCR, Ghaziabad, and Uttar Pradesh, I have 27 years of Teaching experience. He guided 3 PhD students and awarded, 63 M.Pharm students guided. He has Published 10 Indian patents and 08 Indian Design Patent Grant, 08 UK Design patent grants, and 04 Design patent grants are under process. Published 184 Research articles published in National and International journals and 6 Books/Book chapters published. Dr. NGR RAO have a keen interest in research activities as well and always believes in promoting the implementation of newer technological approaches on concept-based learning and teaching in my noble Pharmacy Profession.

    Objective: The aim of this present research work, the aim was to develop drug-loaded Bisoprolol fumarate quick-dissolving tablets by direct compression method by using Trigonella Foenum - Plantago ovata seeds mucilage. Comparative studies between synthetic and natural superdisintegrants. Bisoprolol Fumarate is an antihypertensive drug incorporated in handling hypertension, and as a prophylaxis treatment of angina pectoris, heart failure. Methods: Bisoprolol fumarate-loaded tablets were developed, estimated for different pre and post-compressional parameters, FTIR study, and short-term stability studies. Results and Discussions: The FTIR spectral analysis results indicate no interaction between the drug and formulation additives. pre and post-compressional parameters were within the authorised Pharmacopeial limits, and the results were satisfactory. The disintegration time and dissolution investigations of formulations F12 and F15 were determined to be the most promising, with disintegration times of 22 and 24 secs, respectively. FormulationsF12 and F15 containing Trigonella Foenum and plantago ovata mucilage’s showed highest drug release above 99% within 20 and 25 min respectively. Trigonella Foenum mucilage and Plantago ovata was identified to be the better superdisintegrant compare to synthetic superdisintegrants. Stability studies to optimize the formulations were carried out for about 90 days. During the studies, no considerable changes were noticed in the compressed tablets for in-vitro disintegration time, hardness, friability and uniform distribution of drug content etc. Conclusion: Based on disintegration time the developed formulations F3, F12 and F15 were found to be mostly constitutive, with disintegration times of 30, 22, and 24 secs. The result outcome can be interpreted that the formulations loaded with Bisoprolol fumarate have shown enhanced dissolution rates that might have been the cause of improved bioavailability, subsequently an effective therapy by coupling with Trigonella foenum and Plantago ovata mucilage. These mucilages were plant-derived, non-toxic, bio-compatible, bio-degradable, and least side effects.
  • A review on various formulation methods in preparing Colon targeted mini-tablets for chronotherapy

    Rheumatoid arthritis disease, according to its circadian rhythms shows early morning peak symptoms. Sometimes, single unit (ex. larger tablets) and multiple units (ex. granules, pellets) colon targeted drug delivery systems are not always an efficient treatment option. Because single unit larger tablets may suffer from the disadvantages of unintentional disintegration of the formulation due to manufacturing deficiency or GI variation leading to complete dose dumping. Even the multiple unit drug delivery systems such as granules and pellets also have many drawbacks because of their irregular weights, shapes and sizes. Thus, a tight, reproducible in-vitro and in-vivo release profile can’t be achieved. In an attempt to overcome the problems presented by these delivery systems, advanced system such as multiple unit mini-tablets have developed. This is an approach towards achieving critical factors such as better patient compliance and convenience. In this review, a concerted try has made to provide details of different formulation methods used in preparing mini-tablets of lornoxicam and naproxen drugs for colon targeted delivery in chronotherapy. The techniques formulated and evaluated as core-mini-tablets filled pulsincap drug delivery system (using time dependent polymers), matrix-mini-tablets filled capsule drug delivery system (using microsomal enzyme dependent and pH dependent polymers) and coated-mini-tablets filled capsule drug delivery system (using pH dependent polymers). All these methods were successful in targeting Anti-inflammatory drugs at colonic junction. Hence, the mentioned formulation methods can be successfully used in the chronotherapeutic treatment of Rheumatoid arthritis.
  • Design and development of simvastatin gas powered system for controlled release

    Simvastatin is a Hypolipidemic used to control elevated cholesterol, or hypercholesterolemia. The primary uses of simvastatin are for the treatment of dyslipidemia and the prevention of cardiovascular disease. Gas powered Floating tablets of Simvastatin were developed by direct compression method using different grades of HPMC, Carbopol, ethyl cellulose, sodium CMC, sodium alginate, mixture of sodium bicarbonate, citric acid anhydrous as gas generating agents, Citric acid was also used as an antioxidant. The results of Pre-compressional parameters were within I.P prescribed limits. The prepared tablets were subjected to post compression analysis for the parameters such as hardness, friability, weight variation, thickness, diameter, drug content, lag time subsequently buoyancy time, and in-vitro dissolution studies. Tablets of all the formulation floated more than 12hrs. The results of in vitro buoyancy time and lag time study revealed that as the concentration of sodium bicarbonate increases there is increase in total buoyancy time and decrease in lag time. The results of dissolution study reveals that, a decrease in release rate of the drug was observed on increasing the polymer ratio and also by increasing the viscosity grades of the polymer. In all the formulations, hardness test indicated good mechanical strength, friability is less than 1%, indicated that tablets had a good mechanical resistance. DSC and FT-IR studies revealed that, there was no incompatibility of the drug with the excipients used. The stability study conducted as per the ICH guidelines and the formulations were found to be stable. From this study, it can be concluded that, the formulation retained for longer periods of time in the stomach and provides controlled release of the drug and may improved bioavailability.

    This research work aims to determine the pharmacokinetic parameters and in vitro-in vivo correlation of the selected ileocolonic-targeted coated mini-tablet filled capsule formulation of naproxen. The pure suspension and coated mini-tablet filled capsule formulation of naproxen were administered to adult albino rabbits through the oral route. The samples were analyzed for naproxen by an HPLC method. For the pure drug suspension, the peak plasma concentration was found as 8.499±0.029 μg/ml at 1.139±0.010 hours and the half-life was found to be 9.459±0.387 hours, whereas for the formulation the peak plasma concentration was found as 6.814±0.037 μg/ml at 8.042±0.069 hours and the half-life was found to be 19.657±0.359 hours. This decreased the peak plasma concentration at a delayed time and increased the half-life of the capsule formulation in comparison with the pure drug suspension which showed that naproxen was only targeted to the ileocolonic region. A significant in vitro-in vivo correlation (i.e. R2=0.9901) was also obtained. Thus, the results of these findings suggest that naproxen formulated as coated mini-tablets can be suitable for targeted ileocolonic drug delivery.
