    The chalcone basic skeleton is a unique template which is associated with widespread biological activities. In the present study, a series of novel bichalcones linked with a 1,4-dimethylenepiperazine moiety was prepared through Mannich reaction and Clasien-Schmidt condensation. The synthetic analogs 2-16 were subjected into the cytotoxicity examinations using a panel of 25 human tumor cell lines. Among the tested compounds, 3 and 4 which possessed the 3-pyridyl and phenyl groups as the substructure, respectively, displayed significant cytotoxicity against all the tumor cell lines. The results suggested that these bichalcones were potential to be the anticancer lead drugs.

    A new acylated labdane diterpenoid, 14-deoxy-3,19-diacetyl-11,12-didehydroandrographolide (1), together with three known labdane diterpenoids, wightionolide (2), andrographolide (3) and neoandrographolide (4), and three known flavones, echioidinin (5), skullcapflavone I 2′-methyl ether (6) and echioidin (7), were isolated from the whole plant of Andrographis wightiana. The structure of compound 1 was elucidated by 1D and extensive 2D-NMR spectral studies.

  • Bioactive constituents from the roots of panax japonicus var. major and development of a LC-MS/MS method for distinguishing between natural and artifactual compounds

    Two new saponins, panajaponol (1) and pseudoginsenoside RT1 butyl ester (2), together with 35 known compounds (3-37), were isolated from the roots of Panax japonicus var. major. The structures of 1 and 2 were elucidated on the basis of spectroscopic analysis and chemical methods. Furthermore, a LC-MS/MS method was developed for confirming 2, 3, and 8 as natural compounds containing a butyl ester group. This method should be useful for distinguishing between minor natural and artifactual compounds in Panax species. Moreover, compounds 3, 6, 8, 9, 11, 13, and 15 exhibited strong inhibition of superoxide anion generation and elastase release by human neutrophils in response to formyl-l-methionyl-l-leucyl-l-phenylalanine/cytochalasin B (fMLP/CB), with IC(50) values ranging from 0.78 to 43.6 μM. In addition, 1 showed greater than 2- to 3-fold selective cytotoxic activity against KB and DU145 cancer cell lines.

The structures of these compounds were determined by extensive spectral studies.

    Four new furanoditerpenoids, fibrauretin A ( 1), fibrauretinoside A ( 2), epi-fibrauretinoside A ( 3), and epi-12-palmatoside G ( 4), and a new ecdysteroid glucoside, fibraurecdyside A ( 5), together with seven known compounds including two furanoditerpenoids ( 6 and 7), an ecdysteroid ( 8), and four quaternary protoberberine alkaloids ( 9- 12) were isolated from the stems of Fibraurea tinctoria. The structures of 1- 5 were established on the basis of spectroscopic evidence. Among these compounds, palmatine ( 9) and jatrorrhizine ( 10) showed inhibitory effects against cytochrome P450 3A4 (CYP3A4) with IC 50 values of 0.9 and 2.1 microM, respectively.

  • A new ent-trachylobane diterpenoid from the roots of Sapium sebiferum

    A new ent-trachylobane diterpenoid, saposebifeic acid, together with thirteen known compounds, were isolated from the roots of Sapium sebiferum. Compounds, 5,7,8-trimethoxycoumarin, baccatin, n-alkyl trans-ferulate and 2,6-dimethoxyquinone, were reported for the first time from this plant. The structures of the new and known compounds were established on the basis of extensive 1D and 2D NMR spectral data.

    Three new 2'-oxygenated flavonoids, (2S)-5,7,2',3',4'-pentamethoxyflavanone (1), 5-hydroxy-7,8,2',5'-tetramethoxyflavone (2), and echioidinin 2'-O-beta-d-(6' '-O-acetyl) glucopyranoside (3), together with four known flavonoids, 7-O-methyldihydrowogonin, 7-O-methylwogonin, skullcapflavone I 2'-methyl ether, and skullcapflavone I, and two diterpenoids, andrograpanin and 14-deoxy-11,12-didehydroandrographolide, were isolated from the whole plant of Andrographis affinis. The structures were elucidated by spectral and chemical studies.

    Two new flavonoids, 2',4',6',2,3,4-hexamethoxychalcone (1) and 5-hydroxy-7,2',5'-trimethoxyflavone (2) together with a known flavone glycoside, echioidinin 5-O-beta-D-glucopyranoside (3) were isolated from the whole plant of Andrographis neesiana, and the structures were elucidated by electrospray ionization tandem mass spectrometry (ESI-MS/MS) and one- and two-dimensional (1D- and 2D)-NMR spectral studies including 1H-1H correlation spectroscopy (COSY), heteronuclear single quantum coherence (HSQC), heteronuclear multiple bond connectivity (HMBC) and nuclear Overhauser enhancement spectroscopy (NOESY) experiments.
