Bioanalytical Method Development and its Validation by Using RP-HPLC for Determination of Trimetazidine Dihydrochloride in Human Plasma Sahebrao S. Boraste Boraste, Deelip V. Derle Research Journal of Pharmacy and Technology, 2025 Trimetazidine dihydrochloride is used in the treatment of chest pain in angina condition. The study was performed to determine the bioavailability/bioequivalence of Trimetazidine in human plasma by utilising caffeine as an internal standard. Percent recoveries were obtained as 95.80 % w/v. for trimetazidine. The Liquid-Liquid extraction (LLE) technique was established for the optimal extraction of Trimetazidine and caffeine from biological matrix. The method involved chromatographic separation on Nucleosil C18 (150×4.6mm, 5µm) with mobile phase water, methanol, acetonitrile, and SHS buffer (60:40:10:03 v/v/v/v) at flow rate of 1.3 mL/min with 250C as ambient temperature and injection volume of 5µL. Quantitation was obtained using UV detection at 210 nm with retention times of 3.97 and 13.07 minutes for caffeine and Trimetazidine in plasma respectively. By applying statistics and performing recovery studies the method was validated showing a linearity concentration range of 5.04 - 201.60 ng/mL and a lower limit of quantification of 0.5 ng/mL. The method proved to be rapid, accurate, simple and precise, providing useful data for therapeutic drug monitoring in clinical laboratories and pharmacokinetics study.

Development of In Situ Nasal Gel of Moxifloxacin HCl for Brain Targeting: Design, Optimization and In-vitro Evaluation Khadija Akhlaque Siddiqui, Prashant Lakshaman Pingale, Sahebrao Sampat Boraste, Sunil Vishvnath Amrutkar, Dattatraya Manohar Shinkar Research Journal of Pharmacy and Technology, 2025 Bacterial meningitis is a life-threatening infection of the meninges, often caused by Neisseria meningitidis, Streptococcus pneumoniae, and Haemophilus influenzae type b. Symptoms include high fever, headache, neck stiffness, photophobia, nausea, vomiting, and altered mental status. Diagnosis is confirmed via cerebrospinal fluid analysis, and treatment requires broad-spectrum intravenous antibiotics and corticosteroids. This study aimed to develop a thermoreversible in-situ nasal gel for Moxifloxacin HCl to enhance brain targeting and CNS absorption, reducing enzymatic breakdown and side effects. Using polymers like Xanthan gum, HPMC K4M, and Carbopol-940, the optimized gel demonstrated quick gelling, strong bioadhesion, and high ex-vivo absorption. Stability tests confirmed long-term efficacy. Further in-vivo research is needed to validate these results for meningitis treatment.
Formulation and Evaluation of Transdermal Patches of Clopidogrel Bisulfate Manisha Devram Gangurde, Dattatraya Manohar Shinkar, Sahebrao Sampat Boraste, Prashant Lakshaman Pingale, Sunil Vishvnath Amrutkar Research Journal of Pharmacy and Technology, 2025 Transdermal medication delivery (TDDS) is a viable alternative to oral drug administration, offering benefits such as longer therapeutic efficacy, decreased side effects, enhanced bioavailability, improved patient compliance, and ease of drug therapy termination. A study using solvent casting method prepared nine formulations of transdermal patches using Eudragit L-100 and HPMC K4M as film forming polymers. The patches' weight, thickness, folding endurance, homogeneity of drug content, and moisture content were assessed, along with their organoleptic qualities.in-vitro release, and steady state flux. The optimized transdermal patch (F5) showed the highest drug content of 98.78%, with the highest drug release of 75.91%. The F5 batch showed the highest steady state flux of 2.922 𝝁g/cm² 12hr for clopidogrel bisulfate. The F5 batch completed three months of stability testing, showing no significant alterations to drug content, in vitro drug release, or steady state flux. The findings suggest that patches provide adequate outcomes for longer periods of time, and more clinical research is needed to establish an appropriate transdermal system for hypertension treatment.

Formulation, Development, and Optimization of Cefpodoxime Proxetil Nanosuspension by using 23 Factorial Design Rakesh Sanjay Nath, Sahebrao Sampat Boraste, Dattatraya Manohar Shinkar, Sunil Vishvanath Amrutkar, Prashant Lakshaman Pingale Research Journal of Pharmacy and Technology, 2025 The two main problems with most biologically active compounds are their slow rate of breakdown and their difficulty in dissolving in water. As a member of BCS class IV, cefpodoxime proxetil has limited permeability and solubility. To increase the drug's solubility and rate of dissolution, the current study set out to create Cefpodoxime proxetil nanosuspension utilizing a solvent evaporation technique. Zeta potential, particle size, total drug content, and entrapment efficiency were assessed for the created nanosuspension formulation. Various polymers were used to create stable nanosuspension over various trial batches. Following the findings of trial batches, the polyvinylpyrrolidone k30 was selected using a 23 factorial design for the production of cefpodoxime proxetil nanosuspension. With a %Entrapment efficiency of 96.16±0.55%, batch F4 exhibited the highest percentage among the analyzed formulation batches. Among the batches, the F4 batch has the highest total drug concentration, measuring 95.61±0.32%. Every batch of nanosuspension was assessed for several characteristics; batch F4 exhibited the lowest values for the maximum zeta potential (35.4 mV), polydispersity index (0.042 ± 0.01), and particle size (160.6 ± 1.2nm), in that order. For optimization, a linear model was mostly employed. The results of the ANOVA showed that the F4 batch was the most productive. Polyvinylpyrrolidone K30 and tween 80 were used to create a stable nanosuspension that improved the solubility and subsequent release of pure Cefpodoxime proxetil medication.
