Disruption of hyphal development in Candida albicans ATCC 10231 by bioactive compound from Pseudomonas azotoformans UICC B-91 Yonita Aprilia Putri, Rina Hidayati Pratiwi, Wibowo Mangunwardoyo Journal of Applied Pharmaceutical Science, 2024 Candidiasis and other fungal infections are becoming more frequent. The search for appropriate antibiotics for fungal infections is still ongoing. Pseudomonas azotoformans University of Indonesia Culture Collection (UICC) B-91 has potential as an anticandidal. The purpose of this study was to determine the mechanism of inhibition of the metabolite of P. azotoformans UICC B-91 from the medium against morphologically pathogenic fungi. The results showed metabolite compound P. azotoformans UICC B-91 at a concentration of 100 and 80 mg/ml had strong inhibitory power, and 60 mg/ml had moderate inhibition. Antifungal effect of Pseudomonas azotoformans UICC B-91 metabolites on hyphal growth of Trichophyton mentagrophytes R H Pratiwi, Y A Putri, W Mangunwardoyo, S Sufiandi Iop Conference Series Earth and Environmental Science, 2023 Pseudomonas azotoformans UICC B-91 is known to have antimicrobial potential. The purpose of this study was to determine the inhibition mechanism of the metabolites of P. azotoformans UICC B-91 on the activity of Trichophyton mentagrophytes through morphological observations. The results showed that the metabolites produced by P. azotoformans UICC B-91 were inhibited growth of T. mentagrophytes. The zone of inhibition produced by the extract solution of P. azotoformans UICC B-91 against T. mentagrophytes at concentrations of 100 mg/mL and 80 mg/mL showed strong inhibitory power, with average values of 17.82 ± 0.32 mm and 10.79 ± 0.74 mm, respectively. The microscopic morphology of T. mentagrophytes after treatment with 0.1 g of ethyl acetate extract of P. azotoformans UICC B-91 showed wider hyphae compared to the control hyphae; abnormal cytoplasm; and granular structures between the hyphae, attached to the lateral side of the hyphae, and between the cell wall and the cell membrane. The mechanism of inhibition of the metabolites from the ethyl acetate extract of P. azotoformans UICC B-91 against T. mentagrophytes is thought to involve interference with cell walls and the function of the cell membrane.
Antimicrobial Compound from Endophytic Pseudomonas azotoformans UICC B-91 of Neesia altissima (Malvaceae) Rina Hidayati Pratiwi, Eva Oktarina, Wibowo Mangunwardoyo, Iman Hidayat, Endang Saepudin Pharmacognosy Journal, 2022 P. azotoformans strain UICC B-91 was isolated from Nessia altissima Blume, traditional medicinal plant endemic from Indonesia1 that effective against gonorrhoeae2 due to Neisseria gonorrhoeae infection. But there is no publication research on bioactive natural compound from N. altissma, yet. Endophytic microorganism has genetic diversity due to adaption to host environment and might be transfer gen from host, that relate to the bioactive compound produced.3 Previous research has been observed the antimicrobial activity from centrifugated extract of endophytic P. azotoformans, against several bacteria and C. albicans.4 with notice result on its anticandidal activity. For further observation, the antimicrobial compounds were identified by mass spectrophotometry.
In vitro antimicrobial activities of several extracts endophytic Pseudomonas azotoformans UICC B-91 E Oktarina, R H Pratiwi, W Mangunwardoyo, I Hidayat, E Saepudin Iop Conference Series Earth and Environmental Science, 2021 Pseudomonas azotoformans known for the benefit on agriculture area as soil fertility enhancer. Pseudomonas azotoformans UICC B-91 was successfully isolated from tissues of N. altissima which traditionally known for treating infectious diseases. Therefore, endophytic P. azotoformans might contain antimicrobial compound. This research aims on exploring the antimicrobial activities of several extracts P. azotoformans UICC B-91 against pathogenic microbes. Isolates were extracted with dichloromethane, chloroform, and ethyl acetate. Each crude extract was observed for its potential antimicrobial activity with disc diffusion method against several pathogen microbes. Bioactive compound was assayed by thin layer chromatography (TLC) and screened by bioautography. Results indicated that all crude extract have inhibitory effect on Escherichia coli ATCC 8739, Bacillus cereus ATCC 10876, Staphylococcus aureus ATCC 6583, Staphylococcus aureus ATCC 25923, Salmonella typhimurium ATCC 25241, Pseudomonas aeruginosa ATCC 15442, Bacillus subtilis ATCC 19659 and Candida albicans ATCC 10231. Highest clear zone was on dichloromethane extract. TLC fraction of dichloromethane extract was able to inhibit growth on pathogenic bacteria (except for B. subtilis ATCC 19659 and S. aureus ATCC 6583) and fungi at minimum concentration in the range 125-1,000 μg/mL and 125 μg/mL, respectively. Bioactive compound from endophytic P. azotoformans extracted with dichloromethane have antibacterial and anticandidal effect.
Isolation and structure elucidation of phenazine derivative from Streptomyces sp. strain UICC B-92 isolated from neesia altissima (malvaceae) Rina Hidayati Pratiwi, Iman Hidayat, Muhammad Hanafi, Wibowo Mangunwardoyo Iranian Journal of Microbiology, 2020 Background and Objectives: Endophytic actinomycetes have been known as a promising source for new antibiotics discov- ery against susceptible and resistant forms of pathogenic microorganisms. This study was aimed at determining antibacterial compound from Streptomyces sp. strain B-92 isolated from a medicinal plant Neesia altissima.Materials and Methods: Streptomyces sp. strain UICC B-92 was endophytic actinomycetes of N. altissima that obtained from Universitas Indonesia Culture Collection (UICC). Isolation and determination of bioactive compound were carried out using thin layer chromatography (TLC), nuclear magnetic resonance spectroscopy (NMR), and liquid chromatography mass spectrometry (LC-MS) analyses. An in vitro antibacterial assay of pure bioactive compound from the endophytic actinomycetes strain was performed against Bacillus cereus strain ATCC 10876, Escherichia coli strain ATCC 25922, Salmonella typhimurium strain ATCC 25241, Shigella flexneri strain ATCC 12022 and Staphylococcus aureus strain ATCC25923.Results: The bioactive compound was identified as 4-((3S,4R,5S)-3,4,5-trihydroxy-6-(hydroxymethyl) tetrahydro-2H- pyran-2-yloxy) phenazine-1-carboxylic acid. In vitro antimicrobial assay showed that bioactive compound of Streptomyces sp. strain UICC B-92 exhibited antagonistic activities against two Gram-positive bacteria, viz, B. cereus strain ATCC 10876 and S. aureus strain ATCC 25923.Conclusion: The findings of this research showed that, bioactive compound of Streptomyces sp. strain UICC B-92 is sug- gested a new compound based on glycoside structure and its position.
Identification and screening of rare actinomycetes isolated from Neesia altissima Bl. R. H. Pratiwi, I. Hidayat, M. Hanafi, W. Mangunwardoyo Aip Conference Proceedings, 2017 Actinomycetes is the main source of antibiotics and endophytic actinomycetes from medicinal plants has considerable potential as like the host. The aim of this research is to identify rare actinomycetes isolated from Neesia altissima and to screen their antagonistic activity against diarrhea-causing bacteria in order to find new potential secondary metabolites. Antibacterial compound produced by Pseudomonas aeruginosa strain UICC B-40, an endophytic bacterium isolated from Neesia altissima Rina Hidayati Pratiwi, Iman Hidayat, Muhammad Hanafi, Wibowo Mangunwardoyo Journal of Microbiology, 2017 This study's aim was to determine the identity of antibacterial compounds produced by Pseudomonas aeruginosa strain UICC B-40 and describe the antibacterial compounds' mechanisms of action for damaging pathogenic bacteria cells. Isolation and identification of the compounds were carried out using thin layer chromatography (TLC), nuclear magnetic resonance (NMR) spectroscopy and liquid chromatography mass spectrometry (LC-MS) analyses. Antibacterial activity was assayed via minimum inhibitory concentration (MIC) and the antibacterial compound mechanism was observed morphologically through scanning electron microscopy (SEM). This study successfully identified the (2E,5E)-phenyltetradeca-2,5-dienoate antibacterial compound (molecular weight 300 g/mol), composed of a phenolic ester, fatty acid and long chain of aliphatic group structures. MIC values for this compound were determined at 62.5 μg/ml against Staphylococcus aureus strain ATCC 25923. The mechanism of the compound involved breaking down the bacterial cell walls through the lysis process. The (2E,5E)-phenyltetradeca-2,5-dienoate compound exhibited inhibitory activity on the growth of Gram-positive bacteria.
Identification and characterization of three endophytic bacteria from Neesia altissima (Malvaceae) antagonistic to diarrhea-causing bacteria Pratiwi, R. H., Hidayat, I., Hanafi, M., Mangunwardoyo, W. Malaysian Journal of Microbiology, 2016 Aims: Diarrheal disease is one of serious healthcare problems in developing countries. Endophytic bacteria have been known as a promising source of new antibiotics against susceptible and resistant forms of microorganisms. In this study, we identified three endophytic bacteria isolated from Neesia altissima and screened their antagonistic activity against diarrhea-causing bacteria in order to find new potential secondary metabolites. Methodology and results: Samples of N. altissima were collected from mount Halimun-Salak national park. Endophytic bacteria were isolated from roots, barks, and fresh leaves of N. altissima by surface sterilized method. Screening of antagonistics activity was conducted against five diarrhea-causing bacteria such as Escherichia coli, Staphylococcus aureus, Salmonella typhimurium, Shigella flexneri, and Bacillus cereus using crude extract dilution and diffusion disc methods. Three endophytic bacteria showed antagonistic activity against the pathogenic bacteria. Identification of the three potential endophytic bacteria using molecular analysis showed that two isolates determined as Pseudomonas aeruginosa and one isolate belongs to P. azotoformans. Conclusion, significance and impact study: Crude extract of P. aeruginosa (strain 1.4.1A and 1.2.7D) and P. azotoformans (strain 1.8.7KB) showed growth inhibition activity to the diarrhea-causing bacteria. This is the first report of P. azotoformans exhibited antagonistic activities against diarrhea-causing bacteria. This data suggested that bacterial endophytes from N. altissima provided potential sources for the discovery of new secondary metabolites to combat the diarrhea-causing bacteria. This finding highlights potential prospects of endophytic bacteria utilization from endemic medicinal plants for the discovery of novel bioactive compounds.
