DOSE DEPENDENT IMUNOMODULATORY EFFECTS OF THE HERBAL COMBINATION SAMBILOTO–GINGER–TURMERIC (SIJAKUN): AN IN VIVO STUDY ON WISTAR RATS HANDAYANI, HOTIMAH MASDAN SALIM, ARIFA MUSTIKA, AIN DAROJAH SIDDIQ RAMADHANA, RETNO DIAH PUTRI EKAYANTI International Journal of Applied Pharmaceutics, 2026 Objective: This study aimed to evaluate the effects of Sijakun at different doses on key inflammatory parameters—Neutrophil-to-Lymphocyte Ratio (NLR), macrophage count, TNF-α, and IL-6—in an LPS-induced inflammatory model. Methods: An experimental study was conducted using animal subjects randomly allocated into five groups: negative control, LPS-induced positive control, and three treatment groups receiving Sijakun at graded doses. Inflammatory markers were assessed post-intervention. Data were analyzed using ANOVA for normally distributed parameters and Kruskal–Wallis for non-normal data, followed by appropriate post hoc tests. Results: Sijakun administration significantly influenced all measured parameters (p<0.05). The highest dose (500 mg/kg BW) most effectively reduced NLR, TNF-α, and IL-6 levels, bringing them close to baseline control values. In contrast, the medium dose (250 mg/kg BW) optimally increased macrophage count, indicating enhanced innate immune response. Conclusion: Sijakun exerts significant anti-inflammatory and immunomodulatory effects in a dose-dependent manner. A high dose (500 mg/kg BW) is most effective in suppressing systemic and cytokine-mediated inflammation, while a medium dose (250 mg/kg BW) best enhances macrophage-mediated phagocytosis. These findings support the potential of Sijakun as a complementary herbal agent for modulating LPS-induced inflammatory responses.
Ultrasound-Based Knee Osteoarthritis Severity Assessment and Its Association With Kellgren–Lawrence Grading Rita Vivera Pane, Handayani, Aisyah, Nurhasyimah Hisamud-Din, Aufar Zimamuz Zaman Al Hajiri, et al. Scientific World Journal, 2026 Background Kellgren–Lawrence (KL) grading is a conventional radiography (CR)‐based system commonly used to assess osteoarthritis (OA). Knee OA is characterized by damage to the femoral cartilage (FC); however, CR cannot directly visualize cartilage integrity. Ultrasound (US) may be superior in detecting FC damage. This study is aimed at evaluating the US‐based knee OA severity assessment and analyze its association with KL grading. Methods This was an analytical observational study with a cross‐sectional design which included knee OA patients. All participants underwent knee US scanning by two independent raters. The severity of knee OA was assessed based on three parameters: the contour of the femoral cartilage, the presence of osteophytes, and meniscus protrusion. The results were accumulated and interpreted into grades from 0 to 4. Association between US‐based knee OA assessment and KL grading was analyzed statistically. Results A total of 112 knee OA patients were included in this study. The average age of participants was 61.4 years, with a higher female prevalence. Grade 3 was the most frequently recorded in both US and KL grades. The US interpretation from both raters showed a perfect agreement based on weighted kappa analysis ( κ = 1.00). A chi‐square test showed a significant association between the US and KL grade ( p < 0.001), with a very strong association based on the Cramér′s V test of 0.742 (95% CI: 0.599–0.824). Conclusion This study demonstrates the US‐based knee OA severity assessment and was associated with KL grade.
Effect of Bay Leaf (Syzygium polyanthum) Extract on Antioxidant Activity, MDA Levels, and Liver Histopathology Feature of Ethambutol Induced Wistar Rats Bambang Edi Suwito, Lea Maera Shanty, Retna Gumilang, Handayani Handayani, Renata Alya Ulhaq Indonesian Journal of Medical Laboratory Science and Technology, 2022 Bay leaf extract (Syzygiun polianthum) is one herbal elemnt that may be used to lessen liver function issues, lessen symptops of nausea, vomiting, discomfort and improve adherence and the effectiveness of tuberculossi treatment. The objective of this study was to determine the effect of bay leaf (Syzygium polyanthum) extract on antioxidant activity, Malondialdehyde (MDA) levels, and liver histopathology of ethambutol-induced Wistar rats. This research is a laboratory experiment. Antioxidant activity of Bay Leaf Extract was evaluated by comparing vitamin C with spectrophotometry methods. White Wistar rats were separated into 6 groups and uset to test the lebel of MDA and livers histopathology. Group 1 serves as the control group and received DMSO (placebo); Group 2 was received ethambutol 50mg/kg BW; Group 3 was received ethambutol and silymarine with dose 50mh/kg BW. Group 4-6 had been given ethambutol and extra ethanol extract of bay leaves of 75,150, dan 300mg/kg BW, respectively. The results of the spectrophotometry showed that the Bay Leaf Extract had antioxidant activity comparable to that of vitamin C, with an IC50 of 11.4 g ± (4.4%). One-Way ANOVA test results obtained p = 0.002 (p<0.05) while the Kruskal-Wallis test results obtained the Asymp. Sig value 0.105> 0.05. There was a significance difference in each group’s MDA levels (p=0.002). Although there was no significant difference in the liver histopathology of treated rats (p>0.05). While bay leaf extract significantly lowers MDA levels in ethambutol-induced Wistar rats, it has no discernible impact on the liver histopathology of ethambutol-induced Wistar rats. Bay leaf extract possesses antioxidant activity comparable to vitamin C.
Impact on Medical Education and the Medical Student’s Attitude, Practice, Mental Health, After One Year of the Covid-19 Pandemic in Indonesia Yuda Turana, Putu Asih Primatanti, Wawang S. Frontiers in Education, 2022 IntroductionThe COVID-19 pandemic has caused disruptions in educational institutions across the country, prompting medical schools to adopt online learning systems. This study aims to determine impact on medical education and the medical student’s attitude, practice, mental health after 1 year of the Covid-19 pandemic in Indonesia.MethodsThis study utilized a cross-sectional design. An online questionnaire was distributed digitally to 49 medical schools in Indonesia from February–May 2021. A total of 7,949 medical students participated in this study. Sampling was carried out based on a purposive technique whose inclusion criteria were active college students. This research used questionnaires distributed in online version among 49 medical faculties that belong to The Association of Indonesian Private Medical Faculty. Instruments included demographic database, medical education status, experience with medical tele-education, ownership types of electronic devices, availability of technologies, programs of education methods, career plans, attitudes toward pandemic, and the mental health of respondents. Univariate and bivariate statistical analysis was conducted to determine the association of variables. All statistical analyses using (IBM) SPSS version 22.0.ResultsMost of the respondents were female (69.4%), the mean age was 20.9 ± 2.1 years. More than half of the respondents (58.7%) reported that they have adequate skills in using digital devices. Most of them (74%) agreed that e-learning can be implemented in Indonesia. The infrastructure aspects that require attention are Internet access and the type of supporting devices. The pandemic also has an impact on the sustainability of the education program. It was found that 28.1% were experiencing financial problems, 2.1% postponed their education due to this problems. The delay of the education process was 32.6% and 47.5% delays in the clinical education phase. Around 4% student being sick, self-isolation and taking care sick family. the pandemic was found to affect students’ interests and future career plans (34%). The majority of students (52.2%) are concerned that the pandemic will limit their opportunities to become specialists. Nearly 40% of respondents expressed anxiety symptoms about a variety of issues for several days. About a third of respondents feel sad, depressed, and hopeless for a few days.ConclusionThe infrastructure and competency of its users are required for E-learning to be successful. The majority of medical students believe that e-learning can be adopted in Indonesia and that their capacity to use electronic devices is good. However, access to the internet remains a problem. On the other side, the pandemic has disrupted the education process and mental health, with fears of being infected with SARS-CoV-2, the loss of opportunities to apply for specialty training, and the potential for increased financial difficulties among medical students. Our findings can be used to assess the current educational process in medical schools and maximize e-learning as an alternative means of preparing doctors for the future.
Phytochemical screening, antioxidant activity and cytotoxicity assay from noni juice and fermented noni (Morinda citrifolia L.) Handayani, Devyana Dyah Wulandari, Devyani Diah Wulansari Bali Medical Journal, 2022 Introduction: Noni fruit is one of the plants that is often used in traditional medicine as a medicine for high blood pressure, beriberi, smooth urination, inflammation of the gallbladder, colitis, dysentery, constipation, spleen pain, liver disease, diabetes and back pain. This study aimed to analyze the content of bioactive compounds, antioxidant properties and cytotoxic activity of fermented noni juice and noni juice. Method: Identification of the content of bioactive compounds was carried out using Gas Chromatography (GC), antioxidant activity testing using the DPPH method (2,2-Diphenyl-1 picrylhydrazine) and cytotoxic testing using the Brine Shrimp Lethality Test (BSLT) method. Results: The results showed that the content of phenolic compounds contained in noni juice and fermented noni juice had strong antioxidant properties with the IC50 value for noni juice of 22.23 ppm and for fermented noni juice of 13.95 ppm. Meanwhile, the results of the cytotoxic test of noni juice and fermented noni showed that the LC50 values of 33.26 ppm and 2.25 ppm respectively. Conclusion: It can be concluded that both of these materials have high antioxidant activity and potential as anticancer.
Potential therapy of Sambiloto plant (Andrographis paniculata) using multi-compounds analysis Handayani, Wiwik Winarningsih Gaceta Medica De Caracas, 2021 Introduction: Sambiloto plant (Andrographis paniculata) is often used as herbal medicine plant in Indonesia. Previous evidence indicates the use of a whole plant or single-compound approach. Analysis of multi-compounds is needed to determine the therapeutic potential for standardizing herbal medicine to provide a reliable effect. Methods: An exploratory study searching for the active content of A. paniculata was carried out in the Knapsack program. The chemical structure is analyzed computationally using Prediction of Activity Spectra for Substances (PASS) software. The analysis of the mechanism of action of drug molecules was analyzed using the Search Tool for Interacting Chemicals (STITCH) software. Results: The active content of A. paniculata is 46 types, with 5 of them having 6 effects based on chemical bonds and targeting 12 receptor proteins. Five active contents of A. paniculata include andrographidin A, caffeic acid, chlorogenic acid, wogonin 5-glucoside, and cinnamic acid. Analysis of the mechanism of action of A. paniculata based on 12 target proteins from active ingredients using a multi-compound approach shows 6 unique biological processes. Based on the chemical bonds, 5 active contents of A. paniculata have six effects, including anaphylatoxin receptor antagonist, a beta-adrenergic receptor kinase inhibitor, GABA C receptor agonist, G-protein-coupled receptor kinase inhibitor, Aryl hydrocarbon receptor agonist, and Nicotinic alpha6beta3beta4alpha5 receptor antagonist. Conclusion: There is a therapeutic potential of A. paniculata with multi-compounds analysis. A molecular docking analysis is needed to predict the affinity between the ligand (active ingredient) and the target protein.
